| Objective: Through the combination of experimental research and literature review,this study was designed to sort out the Huangdi Neijing and the later generations’ discussion on blood generation,explore the theoretical basis of liver "generating blood gas",analyze the TCM Mechanism of the theory and the related modern biological substances,so as to guide the experimental research,and verify and elaborate the theory through the experimental research results.Methods: literature review and experimental research were both used.Literature review: Combing the explanation of blood generation in "Huangdi Neijing" and the discussion of blood generation by doctors of later generations,discussing the basic knowledge of Chinese medicine on blood generation of the body.For the theoretical part of liver "generating blood gas",around the symbolic connotation of visceral manifestation of liver in Huangdi Neijing,literature review discussed the relationship between liver generating blood,liver storing blood and liver governing growth,and discussed the relationship between TCM theory with the findings of modern research,and guided the experimental research.Experimental research: 66 SD male healthy rats were randomly divided into 6 groups,one cage for each group.After 3 days of conventional feeding in the animal experimental center of the Affiliated Hospital of Chengdu University of traditional Chinese medicine,the blank 28 day group and the blank 42 day group were not treated as the control group.The animal models of spleen deficiency syndrome were made in the 28 day group and the 42 day group by the method of "dietary irregularities" : The model of spleen deficiency syndrome was established by the treatment of feeding every other day with water unlimited;the animal model of liver depression syndrome was made by the method of tail entrapment and enrage in the 28 day group and 42 day group of liver depression syndrome.The front end of the hemostatic forceps was wrapped with 5 circles of single-layer gauze with width of about 2cm and fixed with transparent adhesive tape.The distal part of the tail of the rats was clamped at 1 / 3 and gradually strengthened as is limited to attack other rats with rage.Each rat is enraged continuously for 3minutes.The total stimulation time of each cage rat is 33 minutes,4 times a day,with an interval of 4 hours.After the model was established for the expected time,the sugar water preference percentage,peripheral blood routine and the content of cytokines EPO,GM-CSF,SCF,IL-3 and TPO were detected.Statistical method: the measured data are expressed as mean ± standard deviation.For those with the same variance,one-way ANOVA is used between groups with LSD test selected for comparison between two groups.For those with the same variance,Welch test is used between groups with Tamhane’s T2 test selected for comparison between two groups.Results:1.The statistical results of blood routine test showed that there was no significant difference in WBC test results between the experimental groups,indicating that the two factors of liver depression and spleen deficiency may not be related to the change of leukocytes.RBC and HGB were significantly different between the blank 42 days group and the liver depression 42 days group(P < 0.05),and the level of red blood cells and hemoglobin content of the former were higher than the latter.PLT count of42 days group of spleen deficiency was higher than that of blank 42 days group(P <0.01).2.There was no significant difference in GM-CSF and IL-3 between the experimental groups,indicating that the factors of liver depression and spleen deficiency might not be related to the changes of these two hematopoietic factors.3.The EPO content of the 28 days group of liver depression was higher than that of the blank 28 days group,the difference was statistically significant(P < 0.05);while the EPO content of the 42 days group of liver depression was lower than that of the blank 42 days group and the 28 days group of liver depression,the difference was statistically significant(P < 0.01);the content of EPO of the 42 days group of spleen deficiency was higher than that of the 42 days group of liver depression(P < 0.05).At the same time,there was no statistical significance between the 28 days group of spleen deficiency,the 42 days group of spleen deficiency and the blank groups with the same modeling time.The EPO content of the blank 28 days group was lower thanthat of the blank 42 days group(P < 0.05).4.The statistical results of SCF showed that: there was no statistical significance between the two blank groups and other experimental groups;the SCF content of the28 days group of liver depression was higher than that of the 42 days group of liver depression with statistical significance(P < 0.01);the SCF content of the 28 days group of liver depression was higher than that of the 28 days group of spleen deficiency with statistical significance(P < 0.05);the SCF content of the 42 days group of liver depression was lower than that of the 42 days group of spleen deficiency,here was statistical significance(P < 0.05).5.The statistical results of TPO showed that the difference between the 42 days group of liver depression and the blank 42 days group and the 42 days group of spleen deficiency was statistically significant(P < 0.05),and the former was less than the latter two.The TPO content of the 42 days group of liver depression was lower than that of the 28 days group of liver depression(P < 0.01);Conclusion:1.In Huangdi Neijing,the visceral manifestation system of the liver is corresponded to spring,which means the function of turning the axis from winter to spring.It has the important function of connecting,turning the pivot still,turning the closed state to the state of moving and dispersing.This function is of great significance to the movement and generation of the body’s Qi and blood.It can promote other viscera to participate in the generation of blood,especially the spleen and kidney.Above is the theoretical connotation of liver "generating blood gas".2.EPO showed an increasing trend in the state of depression,which may be a compensatory increase,so as to have the effect of antidepressant in the early stage of modeling.EPO showed a decreasing trend in depressive state,accompanied by the occurrence of anemia.It is indicated that long-term depression may cause the decrease of EPO,weaken the antidepressant ability,and affect the hematopoietic capacity of the body.3.The change of EPO is related to the state of liver depression,but not to thestate of spleen deficiency.Combined with the previous literature research,it is suggested that EPO may be the important biological basis of "blood gas generation" in traditional Chinese medicine.At the same time,it also has the potential to be used as a judgment index for the classification and grading of depression.4.The effect of chronic unpredictable mild stimulation on EPO may not be related to spleen deficiency syndrome,and can reflect the effect of liver function on EPO.5.With the development of liver depression syndrome,the content of TPO decreased,which may further affect the formation of platelets,indicating that the stagnation of liver qi and the unobstructed drainage may lead to the non accumulation of blood in the liver. |
