Objective:In this experiment,chronic urticaria model rats were used as subjects.By observing Yupingfeng Granules,the expression of IL-23,IL-17 protein,and IL-23,IL-17 m RNA levels in the skin tissue of chronic urticaria model rats were observed.The effects of changes in the skin tissue pathology and its effects on the urticaria based on the IL-23/IL-17 inflammation axis to explore the pathogenesis of the disease and provide experimental basis for the clinical application of Yupingfeng Granules.Method:Set up groups:SPF-grade SD rats were selected,and 60 SD rats were randomly divided into normal group,model group,loratadine group(0.9 mg·kg-1·d-1),Yupingfeng particles high,medium Low-dose group(4.05,2.7,1.3g·kg-1·d-1).The CU rat model was sensitized by injection of ovalbumin with aluminum hydroxide suspension and dipterol vaccine.Hematoxylin-eosin?HE?staining to observe pathological changes of rat skin tissue;Toluidine blue staining to observe degranulation of mast cells in rat skin tissue;immunohistochemical?IHC?method to detect skin tissue IL-23,IL-17 protein Expression;Real-time PCR was used to detect the levels of IL-23 and IL-17 m RNA in skin tissues.Image Pro Plus 6.0 image software was used for semi-quantitative protein IOD scoring,and SPSS 25.0 was used for statistical analysis.Results:Yupingfeng granules can significantly improve the pathological manifestations of dermal edema,widened collagen bundle distance,and inflammatory cell infiltration in urticaria model rats,and can also reduce the degranulation reaction of mast cells in urticaria model rats.Compared with the normal group,the levels of IL-23,IL-17m RNA and protein expression scores in the skin of urticaria rats in the model group were significantly increased?P<0.05,P<0.01?;compared with the model group,Yupingfeng Granules could be significantly down-regulated The level of IL-23m RNA and protein expression scores in the skin of urticaria model rats?P<0.05,P<0.01?,the best effect in the high-dose group;Yupingfeng granules have no significant regulatory effect on IL-17.Conclusion:Yupingfeng granules have the effect of inhibiting inflammatory cell infiltration of skin lesion tissue and restoring tissue structure and morphology of urticaria model rats;Urticaria-like pathological changes,increased mast cell degranulation,and increased expression of IL-23 and IL-17 were observed in the skin tissue of urticaria model rats,suggesting that the IL-23/IL-17 inflammation axis is active in urticaria Plays an important role in the pathogenesis of rats.
