Objective: The purpose of this study was to investigate the expression of Vashibin-1(VASH1),Ubiquitin C-terminal hydrolases L3(UCH-L3)and Hypoxia-inducible factor-1?(HIF-1?)in breast cancer,cancer adjacent tissues as well as benign lesions,analyze the relationship between these proteins and the clinicopathological characteristics of breast cancer,and explore the correlation among VASH1,UCH-L3 and HIF-1?.Methods:Immunohistochemistry(IHC)method was used to detect the expression of VASH1,UCH-L3 and HIF-1? in 97 cases of breast cancer,38 cases of adjacent breast tissues and 29 cases of benign lesions.Results:(1)The positive expression rate of VASH1 in breast cancer cells was lower than that in benign lesions(P<0.05)and in triple negative breast cancer was significantly lower than that in non triple negative breast cancer,and there were significant differences in Luminal types(P<0.05).The expression intensity of VASH1 in breast cancer was weaker than that in adjacent breast tissues and that in benign lesions and was negatively correlated with histological grade and Ki-67,and positively correlated with ER and PR expression(P<0.05).(2)VASH1 was also expressed in the endothelial cells of blood vessels and the intensity of VASH1 expression of endothelial cells in breast cancer was higher than that in adjacent breast tissues and benign lesions(P<0.05).(3)The expression rate of UCH-L3 in breast cancer was significantly lower than that in adjacent breast tissues.Also,The expression rate of UCH-L3 in triple negative breast cancer was significantly lower than that in non triple negative breast cancer(P<0.05).The expression intensity of UCH-L3 in breast cancer was significantly weaker than that in adjacent breast tissues and benign lesions,and was positively correlated with ER and PR(P<0.05).(4)There are two patterns of HIF-1?protein expression: nuclear expression and cytoplasmic expression.The positive rate of nuclei and cytoplasm HIF-1? expression in breast cancer was significantly higher than that in the adjacent breast tissues,and that the expression rate of HIF-1? in the nucleus was significantly higher in breast cancer than that in benign lesions(P<0.05)and higher in the triple negative breast cancer than that in non triple negative breast cancer with the the highest in Erb overexpression breast cancer,followed by basal cell like breast cancer(P<0.05).The expression intensity of HIF-1? in nucleus was negatively correlated with ER and PR,andpositively correlated with Ki-67 and histological grade(P<0.05).On the contrary,the positive expression rate of cytoplasmic HIF-1? in non triple negative breast cancer was higher than that in triple negative breast cancer and the lowest in basal type(P<0.05).The expression intensity of cytoplasmic HIF-1 ? was positively correlated with ER,PR and negatively correlated with Ki-67 and histological grade(P<0.05).(5)In breast cancer,the expression of VASH1,UCH-L3 and cytoplasmic HIF-1? were positively correlated(P<0.05).Conclusions:Lost expression of VASH1 and UCH-L3 and overexpression of HIF-1? might play a role in the oncogenesis and the development of breast cancer.The transformation of HIF-1? from cytoplasm into nucleus indicates the malignant evolution of tumor.VASH1,UCH-L3 and HIF-1? have certain value in the malignant degree evaluation of breast cancer.To increase the expression of VASH1,UCH-L3 and to prevent the migration of HIF-1? from cytoplasm to nucleus may be helpful for the prevention and treatment of breast cancer,especially for triple negative breast cancer.
