Objective:Tumor growth and metastasis are accompanied by a large number of new blood vessels and glucose metabolism.Angiogenesis requires both vascular growth factor and vascular inhibition factor,the most important discovery of vascular growth factor is vascular endothelial growth factor(VEGF),It promotes the formation of new blood vessels by stimulating the proliferation and migration of endothelial cells(ECs).However,Vasohibin(VASH)has recently been discovered as a new angiogenesis inhibitors.It includes VASH1 and VASH2.The increase of glucose metabolism was closely related to the up expression of glucose transporter.There are eight different subtypes of glucose transporter.Among them,glucose transporter 1(Glut-1)is a protein with high glucose capacity in transmembrane transport.In malignant tumor tissue,tumor cells accelerate the transfer of extracellular glucose into the cell by up-regulating the expression of Glut-1.Thus,cell proliferation and tumor growth were enhanced.In this paper,mRNA and protein expression of VASH 1,VEGF and Glut-1 in colorectal cancer tissues and their adjacent normal tissues were detected by qRT-PCR and immunohistochemical(IHC)SP staining.The relationship between the clinicopathological features of colorectal cancer was analyzed to explore its role in the occurrence,development,invasion and metastasis of colorectal cancer.Method:64 clinical specimens taken from Clinical Medical College of Yangzhou University(Northern Jiangsu people's hospital)gastrointestinal surgery in March 2014 to March 2015,collected colorectal cancer tissue and normal tissue(more than 5 cm cancers edge)were divided in two groups.None of the patients had received chemotherapy,radiotherapy or biological treatment before surgery.All specimens were confirmed by postoperative pathology and corresponding clinical case data.All colorectal cancer tissues and adjacent tissues were excised within half an hour after the surgery,and divided into two groups.One group was put in liquid nitrogen,than transfer to-80? refrigerator.The other was fixed in formalin solution,kept in 4? refrigerator.The expression of VASH 1,VEGF and Glut-1 protein in tissue was detected by immunohistochemical staining.qRT-PCR was used to detect the expression of VASH1,VEGF and Glut-1 mRNA.The relationship between the expression of the three and the characteristics of clinical cases was analyzed.Result:The expression of VASH 1,VEGF and Glut-1 in cancer tissues was higher than that of adjacent normal tissues,all these three genes were related to T stage,lymph node metastasis,distant metastasis and pathological stage(P<0.05),yet,it had no relationship with gender,age,tumor size,tumor type,and vascular invasion.VASH1 was positively correlated with VEGF?Glut-1(r=0.9058,p<0.01;r=0.9251,p<0.001).Survival analysis suggested that the positive expression of N stage,TNM stage,VASH1,VEGF and Glut-1 was a risk factor for survival(P<0.05).However,there was no significant difference in gender,age,tumor size,differentiation degree,T stage,M stage and survival rate.Further,the risk factors were included in Cox regression analysis,and TNM staging and VEGF positive expression were independent risk factors affecting survival prognosis(P<0.05).Conclusion:The mRNA and protein expression levels of VASH1,VEGF and Glut-1 in colorectal cancer tissues were higher than those of adjacent normal tissues.VASH1 expression was related to tumor differentiation,TNM stage,T stage,lymph node metastasis and distant metastasis.The expression of VEGF was related to tumor differentiation,TNM stage,T stage,lymph node metastasis and distant metastasis.Glut-1 expression was associated with TNM stage,T stage,lymph node metastasis and distant metastasis.The expression levels of VASH1,VEGF and Glut-1 in colorectal cancer tissues were positively correlated.The positive expression of VASH1,VEGF and Glut-1 is a risk factor affecting survival rate.TNM staging and VEGF expression were independent risk factors for survival prognosis.
