Objective:The aim of this study was to characterize the chemical constituents and screen the potential antioxidative compositions and different components of seabuckthorn oil,providing a reference for the quality evaluation of seabuckthorn oil.Methods:Samples of seabuckthorn pulp oil and seed oil were collected.The chemical components of them were characterized by multi-instrument platforms,and their antioxidant activities were determined.Based on chemometrics methods,the potential antioxidative compositions of seabuckthorn oil and the different components of the two seabuckthorn oils were screened.1.Quantitative determination of 9 fatty acids in seabuckthorn oil by GC-MS:After fatty acid methyl esters were obtained through fatty acids derivatization,they were separated on a DB-17ms column and were measured by MRM mode.Methodology validation was performed;2.Qualitative analysis of volatile components in seabuckthorn oil by HS-GC-MS:Headspace sampling method,a HP-5ms capillary column and full scan mode were used to detect sample,and MS-DIAL software was used for component identification;3.Qualitative analysis of glycerides in seabuckthorn oil by UHPLC-Q-TOF-MS/MS:the BEH C18column and methanol-isopropanol were used as chromatographic column and mobile phase,respectively.The diglycerides and triglycerides of seabuckthorn oil were determinated in ESI positive ion mode;4.Qualitative analysis of seabuckthorn oil ethanolic extract based on UHPLC-Q-TOF-MS/MS combined with molecular networking:A BEH C18column and acetonitrile-water were used as chromatographic column and mobile phase,respectively.The composition of seabuckthorn oil ethanolic extract was determined in ESI negative ion mode,and was identified by molecular networking technology;5.Determination of the antioxidant activity of seabuckthorn oil and its ethanolic extract:DPPH and ABTS method were used to determine the antioxidant activity;6.Screening of potential antioxidative compositions and comprehensive comparison of seabuckthorn seed oil and pulp oil components:Seabuckthorn oil component-activity correlation models were established to screen potential antioxidative compositions based on chemometrics methods.Orthogonal partial least squares-discriminant analysis and t test were used to comprehensively compare the composition of seabuckthorn seed oil and pulp oil to find different components.Results:13 seabuckthorn pulp oil samples and 13 seed oil samples were collected;A GC-MS quantitative method for the determination of 9 fatty acids in seabuckthorn oil was established.The linearity,precision,accuracy and solution stability of the method met the requirements.The contents of 9 fatty acids in each seabuckthorn oil sample were determined;Through HS-GC-MS analysis,77 volatiles in seabuckthorn oil were identified,including esters,alcohols,aldehydes and alkanes;85 glycerides were identified by UHPLC-Q-TOF-MS/MS method,including 9 diglycerides and 76triglycerides;A total of 79 components were identified from ethanolic extracts of seabuckthorn oil,including tocopherols,flavonoids,fatty acids,et al;The experimental results of antioxidant activity showed that the TEAC values of the seabuckthorn oil and its ethanolic extracts measured by the ABTS method were between 3.93.1-2.51 mmol/g and 2.91-10.07 mmol/g,respectively.The IC50determined from DPPH method were between 2.15-18.17 mg/m L and3.57-19.58 mg/m L;Based on the component-antioxidant activity correlation analysis,11 potential antioxidative compositions were finally screened,including?-tocopherol,?-tocotrienol,dehydro-?-tocopherol,?-tocotrienol?or?-tocotrienol?,methyleugenol,ferulic acid,gallocatechin,aragenic acid,isoneriucoumaric acid,4-hydroxybenzoic acid,and palmitoic acid;78different components between pulp oil and seed oil were screened by using chemometrics method,include 21 volatiles,15 glycerides and 42 ethanolic extract components.Conclusion:In this study,the components of seabuckthorn oil were systematically analyzed and the antioxidant activity was determined.11potential antioxidative compositions were screened the.78 different components of seabuckthorn pulp oil and seed oil were identified,of which 7were related to antioxidant activity.This study laid the foundation for the determination of quality control indicators and the establishment of a quality evaluation system for seabuckthorn oil.
