Part 1 Changes of the number of endothelial progenitor cells in peripheral blood of patients with lung adenocarcinomaObjective:1.To detect the expression of endothelial progenitor cells in the peripheral blood in patients with lung adenocarcinoma;2.To observe the changes of EPC expression level after treatment with chemotherapy drugs(pemetrexide+platinum)and chemotherapy drugs(pemetrexide+platinum)combined with anti-angiogenic drugs(bevacizumab);3.To explore whether EPC can be used as one of the predictors of potential lung adenocarcinoma and the effect of anti-angiogenic drugs(bevacizumab)on peripheral blood EPC in patients with lung adenocarcinoma;4.Serum expression levels of VEGFA and VEGFB in normal subjects and patients with lung adenocarcinoma were detected and the correlation between serum expression of VEGFA,VEGFB and EPC in patients with lung adenocarcinoma was analyzed.Methods:1.34 patients with lung adenocarcinoma(among them,primary treatment group: 12 cases;Chemotherapy group: 11 cases;Chemotherapy + bevacar group: 11 cases)and eight healthy controls were collected.CD133,CD34 and VEGFR-2 were all positive and CD45 negative cells were used as surface markers of EPC.2.The remaining blood samples were centrifuged,the upper serum was collected and stored in the refrigerator at-80 ?.The expression levels of VEGFA and VEGFB in each sample were detected by Elisa,and the correlation between these samples and EPC in peripheral blood of patients with lung adenocarcinoma was studied.Results:1.The expression level of EPC in peripheral blood of the initial treatment group was significantly higher than that of the control group(P<0.05);the expression level of EPC in peripheral blood of the chemotherapy group was higher than that of the initial treatment group(P<0.05);while the expression level of EPC in peripheral blood of the chemotherapy + bevacizumab group was lower than that of the chemotherapy group(P<0.05).2.The expression of VEGFA and VEGFB in the serum of patients with lung adenocarcinoma was significantly higher than that of healthy controls(P<0.05);The expression of VEGFA in serum was positively correlated with EPC in peripheral blood of patients with lung adenocarcinoma(r=0.593;P<0.05);The expression of VEGFB in serum was positively correlated with EPC in peripheral blood of patients with lung adenocarcinoma(r=0.440;P<0.05).Conclusions:1.EPC in peripheral blood of lung adenocarcinoma patients may be involved in the formation of pulmonary adenocarcinoma blood vessels.The application of chemotherapy(pmetrexide+platinum)can promote the expression of EPC in peripheral blood of patients.The application of Chemotherapy(pemetrexide+platinum)combined with anti-angiogenic drugs(bevacizumab)can inhibit the mobilization of EPC in peripheral blood of patients,resulting in the reduction of EPC in peripheral blood of patients with lung adenocarcinoma.2.VEGFA and VEGFB promote the expression of EPC in peripheral blood of patients with lung adenocarcinoma,which leads to the formation of tumor blood vessels.Therefore,EPC can be used as one of the predictors of potential lung adenocarcinoma,providing a theoretical basis for early clinical detection of lung adenocarcinoma.Part 2 Bioinformatics analysis of lung adenocarcinomaObjective: 1.To investigate the differential expression of m RNA of related genes(VEGFA,VEGFB)in lung adenocarcinoma patients and normal tissues.2.The correlation between the m RNA expression level of the above factors and the prognosis of patients with lung adenocarcinoma was analyzed.Methods:The oncomine database was used to mine the differential expression of the lung adenocarcinoma related genes(VEGFA,VEGFB)m RNA in cancer tissues and normal tissues,and the online Kaplan-Meier plotter website analysis showed that the expression levels of VEGFA,VEGFB m RNA had a certain influence on the prognosis of lung adenocarcinoma patients.Results: Through oncomine database analysis,we found that the expression levels of VEGFA,VEGFB m RNA in lung adenocarcinoma were higher than that in normal people,and all of them were statistically significant(P<0.05);Through Online Kaplan-Meier plotter website analysis,we found that the expression levels of the above related factors VEGFA and VEGFB m RNA were inversely proportional to the prognosis of lung adenocarcinoma and were statistically significant(P<0.05).Conclusions: Through the analysis of oncomine database,we found that the expression levels of VEGFA and VEGFB m RNA in patients with lung adenocarcinoma were higher than those of normal people,and the higher the expression levels of VEGFA and VEGFB in patients with lung adenocarcinoma,the worse the prognosis was.Therefore,through the conclusion of the two parts,we speculated that patients with lung adenocarcinoma with high expression of VEGFA and VEGFB m RNA can promote the increase of EPC level,and thus promote the formation of blood vessels in patients with lung adenocarcinoma,resulting in poor prognosis of patients with lung adenocarcinoma.
