Objective The differential expression profile of circ RNA in lung adenocarcinoma tissues was analyzed to find a new potential diagnostic molecule.Methods Four pairs of lung adenocarcinoma tissues and adjacent tissues were analyzed by microarray technique,and the obtained circ RNA differential expression profile was preliminarily analyzed.Then,we screened out the significantly differentially expressed circ RNA.Afterwards,the q RT-PCR technique was used to further verify the expression of target circ RNA in 26 pairs of samples.After that,we statistically analyzed the correlation between the expression level of the target circ RNA and clinicopathological features,and its diagnostic value was analyzed by receiver operating characteristic curve.Further,circ RNA-mi RNA-m RNA interaction analysis of the target circ RNA and GO and KEGG analysis of target genes will performed to speculate the possible role of the target circ RNA in lung adenocarcinoma.Results We discovered 168 differentially expressed circRNAs,of which 127 were up-regulated and 41 were down-regulated.Among them,hsacirc RNA000121 is a molecule which significant differentially expressed.And the consequence verified in 26 pairs of samples was consistent with the microarray technique.Its expression level of lung adenocarcinoma tissues was significantly higher than that of adjacent tissues.Statistical analysis showed that the expression level of hsacirc RNA000121 was not related to age,gender and tumor diameter of patients,but was closely associated with lymph node metastasis and a high TNM stage.And its ROC curve area was0.712.Bioinformatic analysis revealed that the target genes of hsacirc RNA000121are related to the basic biological processes such as proteoglycan metabolic process,cell cortex,histone deacetylase complex,protein serine/threonine/tyrosine kinase activity and so on.They are also closely connectted with Rap1 signaling pathway.Conclusion Circ RNA were expressed in lung adenocarcinoma and adjacent tissues,and the significantly different hsacirc RNA000121 is highly expressed in lung adenocarcinoma tissues.Moreover,the expression level of hsacirc RNA000121 may be related to the invasiveness of lung adenocarcinoma.Hsacirc RNA000121 may be a new biomarker for detection of lung adenocarcinoma.
