Objective: To study the expression and methylation status of PGM5-AS1 in laryngeal squamous cell carcinoma cell lines and tissues,to analyze the relationship between its expression,methylation status and clinicopathological features,and to explore the effect of methylation status on the expression.Methods:1.q RT-PCR method was used to detect the expression of PGM5-AS1 in 4 laryngeal squamous cell carcinoma cell lines,48 laryngeal squamous cell carcinoma tissues and corresponding normal tissues.the changes of PGM5-AS1 expression in cell lines before and after 5-Aza-d C treatment were detected.2.BGS method was used to detect the methylation status of PGM5-AS1 in 4 laryngeal squamous cell carcinoma cell lines.3.MSP method was used to detect the methylation status of PGM5-AS1 in laryngeal squamous cell carcinoma cell lines before and after 5-Aza-d C treatment,laryngeal squamous cell carcinoma tissues and corresponding normal tissues.Results:1.The expression of PGM5-AS1 was low in laryngeal squamous cell carcinoma cell lines,and the expression of PGM5-AS1 was significantly increased in laryngeal squamous cell carcinoma cell lines treated with 5-Aza-d C.Compared with normal tissues,the expression in laryngeal squamous cell carcinoma tissues was significantly decreased.The expression level of PGM5-AS1 in tumor tissues was closely related to TNM stage,and the expression in III+ IV stage was significantly lower than that in I+II stage.2.The results of BGS method showed that PGM5-AS1 was hypermethylated in laryngeal squamous cell carcinoma cell lines.3.The results of MSP assay showed that PGM5-AS1 was hypermethylated in laryngeal squamous cell carcinoma lines,and the degree of methylation decreased after 5-Aza-d C treatment.Compared with the normal tissues adjacent to the cancer,there was hypermethylation in laryngeal squamous cell carcinoma tissues.The methylation rate of PGM5-AS1 in laryngeal squamous cell carcinoma tissues was correlated with TNM stage.Compared with the non-methylated group,the expression level of PGM5-AS1 in the methylated group was significantly decreased.Conclusion:1.The low expression of PGM5-AS1 in laryngeal squamous cell carcinoma cell lines and laryngeal squamous cell carcinoma tissues suggests that it may play a role as a tumor suppressor gene.2.The expression level and methylation status of PGM5-AS1 are related to TNM stage,which indicates that it may be related to the malignant degree and progression of laryngeal squamous cell carcinoma.3.Methylation of PGM5-AS1 may be one of the mechanisms leading to the decrease of its expression.
