Objectives:To detect the expression levels of STX3,PTEN and PIK3CA in endometrial cancer,and compare the expression differences of the three in endometrial carcinoma,atypical hyperplastic endometrium and normal endometrium.The correlation between STX3 and various clinicopathological indicators of endometrial cancer and the relationship between STX3 and PTEN and PIK3CA expression were analyzed,and the significance of STX3 in endometrial cancer was discussed.Methods:A total of 87 tissue specimens were collected from the Department of Pathology,First Affiliated Hospital of Jinan University,Guangzhou.They were divided into 46cases in endometrial cancer group,14 cases in atypical hyperplasia group and 27cases in normal endometrium group.Immunohistochemistry and Western blotting were used to detect the expression levels of STX3,PTEN and PIK3CA in each group.The relationship between STX3,PTEN and PIK3CA was analyzed by?2 test or Spearman's correlation.?2 test was used to analyze the correlation between STX3expression and various clinicopathological indexes of endometrial cancer.Results:1.Immunohistochemical and Western blotting results showed that STX3 expression increased in endometrial cancer group,with significant difference?p<0.05?.2.Immunohistochemical and Western blotting results showed that PTEN expression decreased in endometrial cancer group,with significant difference?p<0.05?.3.Immunohistochemical and Western blotting results showed that PIK3CA expression increased in endometrial cancer group,with significant difference?p<0.05?.4.By?2 test or Fisher exact probability and Spearman's correlation analysis,there is a correlation between the expression of STX3,PTEN and PIK3CA in endometrial cancer?p<0.05?,and the correlation coefficients are-0.316 and 0.360 respectively.5.In endometrial cancer group,the expression of STX3 was found to be correlated with age,FIGO stage,tumor differentiation and adnexal metastasis?p<0.05?.There was no significant correlation with menstruation,histological type,myometrial invasion depth,vascular tumor thrombus and lymphatic metastasis?p>0.05?.Conclusion:1.The expression of STX3 in endometrial cancer group is increased.The STX3positive patients are younger,FIGO stage is earlier,tumor differentiation degree is better,and trend to exist accessory metastasis.2.The positive expression rate of STX3 in endometrial cancer tissues was highest in stage I,followed by stage IIIIV,and lower in stage II,which was consistent with the expression of PTEN.3.STX3 has a negative correlation with PTEN expression,while STX3 has a positive correlation with PIK3CA expression.STX3 may be related to PTEN-PI3K/AKT pathway.
