Objective: Through bioinformatics analysis of microarray related to pancreatic duct adenocarcinoma(PDAC),it provides a theoretical basis for studying the molecular mechanism of PDAC occurrence and development,and provides candidate targets for its diagnosis and treatment.Methods: Download microarray data sets from gene expression omnibus(GEO)to determine the differentially expressed genes(DEGs)of PDAC occurrence and development,and perform functional enrichment analysis.The functional annotation,pathway analysis,protein-protein interaction network(PPI)analysis and survival analysis of differentially expressed genes were performed by DAVID online analysis software,STRING online database,Cytoscape software and GEPIA online analysis tool.Further screening to obtain candidate targets,and further analysis of candidate targets to obtain relevant results.Results: A total of 234 DEGS of PDAC were screened,of which 162 were up-regulated and 56 were down-regulated,16 of which showed different expression levels in different microarray data sets,so they were excluded;functional enrichment analysis found that these genes mainly enriched in biological processes such as redox process,inflammatory reaction,negative regulation of apoptosis,PI3K-Akt signaling pathway,amoebiasis,platelet activation,regulation of actin cytoskeleton,amino acid biosynthesis,protein digestion and absorption,pantothenate and coenzymeA biosynthesis.PPI analysis obtained 1 key module and 10 hub genes,namely THY1,COL1A2,COL1A1,FN1,ALB,EGF,ITGA2,CXCR4,HSPG2 and KRT19.Prognostic analysis and disease-free survival showed that high expression of ITGA2 and KRT19 can predict poor survival outcome of PDAC.Conclusion: This study identified hub genes ITGA2,KRT19,and key pathway PI3K-Akt signaling pathway of PDAC,which may be potential targets for its diagnosis,treatment,and prognosis.
