Objective: Breast cancer is one of the most important malignant tumors that threaten women's health.Although there has been some progress in the diagnosis and treatment of breast cancer,the incidence and mortality of breast cancer are still high.This research discusses the effects of prominin2(PROM2)on proliferation,migration and invasion of breast cancer and its underlying mechanisms,and hopes to help find effective prognostic markers and therapeutic targets for breast cancer.Methods: This research analyzed the expression and prognosis correlation of PROM2 in 1085 breast cancer tissues,291 normal breast tissues,and 6 groups of HER2-induced aggressive MCF10 A cells and 6groups of untreated non-invasive MCF10 A cells,and the correlation between PROM2 and PI3K/AKT pathway factors in TCGA,GTEx and GEO databases by bioinformatics.The expression of PROM2 in breast cancer tissues and adjacent tissues were further detected by real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)andimmunohistochemistry(IHC).ZR-75-1,MCF-7,SK-BR-3 breast cancer cell lines and HBL-100 normal breast cells were selected,and the expression of PROM2 in each cell line was detected by qRT-PCR and immunofluorescence(IF).Breast cancer cells with up-regulated and down-regulated PROM2 expression were constructed by plasmid transfection,and the effects of PROM2 on proliferation,migration and invasion of breast cancer cells were examined by MTT,crystal violet,scratch and Transwell assays,and the effect of PROM2 on the PI3K/AKT pathway was detected by IF.Results: Database analysis showed that the expression of PROM2 in breast cancer tissues was higher than that in normal breast tissues,and the higher the expression,the lower the overall survival rate of patients(P<0.01),and PROM2 expression was higher in HER2-induced invasive MCF10 A cells than in untreated non-invasive MCF10 A cells(P<0.05).Furthermore,it was found that PROM2 was significantly positively correlated with the pathway factor of PI3K/AKT pathway(PI3K: R=0.5,P<0.01;AKT: R=0.44,P<0.01).This research also demonstrated that the expression of PROM2 in breast cancer tissues and breast cancer cells are higher than that in adjacent tissues and normal breast cells(P<0.01),and PROM2 can promote the proliferation,migration and invasion of breast cancer cells(** P < 0.01).It was also proved that up-regulation of PROM2 promotes the expression and activation of AKT signals..Conclusion: PROM2 is highly expressed in breast cancer and may promote breast cancer proliferation,migration and invasion through activation of the PI3K/AKT pathway.
