Objective: To analyze the expression of circRNA0102618 in breast cancer tissues and cells,and to investigate the role of circRNA0102618 in the proliferation,migration and invasion of breast cancer cells.Methods: 1.CircRNA 0102618 with significantly different expression in breast cancer and paracancer tissues was screened by circRNA microarray.2.The postoperative breast cancer tissues,paracancer tissues and related clinicopathological data were collected.The expression levels of circRNA0102618 in breast cancer tissues,paracancer tissues,normal breast epithelial cells MCF-10 A and breast cancer cells MCF-7 were quantitatively analyzed by qRT-PCR.The relationship between the expression of circRNA0102618 in breast cancer and the clinicopathological characteristics of patients was analyzed.3.MC-7 cell lines with stable overexpression of circRNA0102618 were constructed by lentivirus transfection.The effects of overexpression of circRNA0102618 on the proliferation,migration and invasion of MCF-7 cells were studied by Plate clone formation assay,CCK8 assay,Wound Healing assay,and Transwell assay.Results: 1.Low expression of circRNA0102618 in breast cancer was screened by circRNA expression profile chip;2.The expression of circRNA0102618 in breast cancer tissues was significantly lower than that in paracancer tissues(P=0.004).The expression of circRNA0102618 in MCF-7 was significantly lower than that in MCF-10A(P=0.009).3.The expression level of circRNA0102618 in the positive lymph node group was significantly lower than that in the positive lymph node group,and the difference was statistically significant(P=0.033).The expression level of circRNA0102618 in the HER-2 positive group was significantly lower than that in the HER-2 negative group(P=0.048).The expression level of circRNA0102618 in CK/5/6 positive group was significantly lower than that in ck5/6 negative group(P=0.009).The expression of circRNA0102618 in non-invasive carcinoma was significantly higher than that in both non-invasive and non-invasive carcinoma(P < 0.05).4.Both the plate cloning experiment and the cck-8 experiment showed that the proliferation capacity of MCF-7 with overexpression of circRNA0102618 was significantly lower than that of the non-transfected group(P < 0.05).The results of both the scratch test and the Transwell test showed that compared with the untreated group,the migration ability and invasive ability of MCF-7 with circRNA0102618 were significantly reduced(P < 0.05).Conclusion: CircRNA0102618 was significantly down-regulated in breast cancer,and its low expression was associated with poor pathological typing,positive lymph node metastasis,overexpression of HER-2 and overexpression ofCK5/6.Overexpression of circRNA0102618 can inhibit the proliferation,invasion and other biological functions of McF-7 in breast cancer cells.
