| Aim:We measured the expression pattern of serum circ RNA in the progression of colorectal polyp-colorectal cancer by Arraystar Human circ RNA Array to understand the dynamic expression pattern of serum circ RNA in colorectal polyp malignant transformation.Furthermore,potential candidate circ RNAs were analyzed to lay the foundation for exploring the development and mechanism in colorectal polyp-colorectal cancer.Methods:1.We collected 10 samples of blood samples from healthy individuals,colorectal polyps,and colorectal cancer patients.Among them,5 cases of which were analyzed by Human circ RNA Array,and 5 cases were verificated by q RT-PCR.2.The differential expression profiles of circ RNA in 15 serum samples were analyzed by Human circ RNA Array.And then we further analyzed the differential expression pattern of circ RNA in healthy control-polyp group,polyp-intestinal cancer group,healthy control-colorectal cancer group,and screened out 10 Circ RNAs with significant differences.3.q RT-PCR was used to dected the expression of the above 10 differentially expressed circ RNAs in 5 cases of healthy individuals,colorectal polyps,and colorectal cancer patients to verify the reliability of the chip results.4.We also predicted the interaction between the 10 candidate circ RNA with mi RNA based on Target Scan & mi Randa database.Results:1.Totally 2162 differential expression of human circ RNAs was altered in peripheral circulation of CRP and CRC patients compared to healthy individuals by Human circ RNA Array.We found that there were 52 circ RNAs upregulated,while 38 circ RNAs downregulated in CRP group compared with healthy group.64 circ RNAs were upregulated,while 72 circ RNAs were downregulated in CRC group compared with healthy group.And 38 circ RNAs were upregulated,while 81 circ RNAs were downregulated in CRC group compared with CRP group.2.we screened 5 circ RNAs from normal to polyp expression downregulated and polyp to colon cancer expression upregulated and 33 circ RNAs from normal to polyp expression upregulated,polyp to colon cancer expression downregulated.3.These 10 circ RNAs including circ0001013,hsacirc0001907,hsacirc0005758,hsacirc0029431,hsahsacirc0000669,hsacirc0046188,hsacirc0053063,hsacirc0007422,hsacirc0003915 and hsacirc0007883 were selected out.And then the m RNA expression levels of 4 potential candidate circ RNAs,hsacirc0001013,hsacirc0005758,hsacirc0007422 and hsacirc0053063 showed an upregulated and then downregulated trend in healthy control group,polyp group and colorectal cancer group,which was consistent with the Human circ RNA Array analysis.However,the m RNA expression levels of hsacirc0053063 showed significant differences between the polyp group and the colorectal cancer group.4.The interaction between the differential expression of candidate hsacirc0001013,hsacirc0005758,hsacirc0007422,hsacirc0053063 and micro RNA was further predicted.Conclusion:We obtained the circ RNA expression profile of healthy individuals,colorectal polyps,and colorectal cancer patients and screening potential candidate circ RNAs,hsacirc0001013,hsacirc0005758,hsacirc0007422,provides new directions for the research development and mechanism in normal-polyp-colorectal cancer. |
