Objective:To probe into the influence of basic fibroblast growth factor(bFGF)on the expression of Bcl-2 and Bax in neonatal rats with hyperbilirubinemia.Methods:Totally 120 7-day-old neonatal SD rats were selected and randomly divided into 3groups according to nest and weight: normal contral group(A group n=40),hyperbilirubinemia group(B group n=40),basic fibroblast growth factor interwention group(C group n=40).The hyperbilirubinemia model was established by intraperitoneal injection of bilirubin solution,intervention with intrapertioneal injection of bFGF immediately after C group model was established.A group was injected with 0.9%sodium chloride injection of the same volume.Rats were executed at 6,12,24,48 and 72 hours.HE staining,immunohistochemistry and Tunnel method were used to measure the expression of Bcl-2 and Bax in hippocampus and the results of apoptosis.Results:(1)By observing the neural behavior of rats: There was no abnormal neural behavior in group A,and there were different degrees of abnormal neural behavior(such as fidgety,decreased activity,weak escape response to external stimulation,prostration,etc)in group B and C.(2)In group A,there was no or only a few neurocyte apoptosis in hippocampus,and in group B and C,the number of neurocyte apoptosis were more than that in group A,and that in group B was more than that in group C.In group B and C,the expression of Bcl-2 and Bax was higher than that in group A,the expression of Bcl-2 was significantly increased in group C,and Bax was significantly increased in group B,the difference was statistically significant(P<0.05).Conclusion:(1)7-day-old SD rats which suffer from hyperbilirubinemia may have nervous system damage.(2)The apoptosis of neurona in the hippocampus of 7-day-old SD rats with hyperbilirubinemia may be related to Bcl-2 and Bax.After bFGF interwention,the apoptosis of neurons may be reduced by increasing the expression of Bcl-2 and reducing the expression of Bax.It indicated that bFGF could reduce bilirubin-induced brain injury and protect nerve cells to certain extent.
