Objective To investigate the effects and mechanisms of different doses of Lycium barbarum polysaccharides?LBP?on Wnt/?-catenin signaling pathway in renal interstitial fibrosis in rats with unilateral ureteral obstruction?UUO?.Methods The RIF rat model was established by unilateral ureteral obstruction?UUO?.108 SPF SD healthy adult male rats were randomly divided into sham operation group,model control group and treatment group.The treatment group was divided into LBP low,medium and high-dose group and Benner Pury group.18 rats were in each group.After 1 days of continuous gavage for 3 weeks,LBP low,medium and high dose groups were given 400mg?kg·D?-1,600 mg?kg·D?-1,800mg?kg·D?-1.Benner Pury group was given 1.05mg hydrochloride?kg·D?-11 intragastric administration,sham operation group and model control group were daily fed normal saline solution.Six rats were randomly sacrificed on the 7,14 and 21st days after modeling to test the serum creatinine?Scr?level and calculate the kidney organ index;the operational side kidney tissue respectively by HE staining and Masson staining were related to pathological examination;the expression of Wnt4,?-catenin and TGF-?1 in renal tissues was detected by immunohistochemistry and Real Time PCR.Results?1?compared with the sham operation group,the serum Scr and kidney index of model group LBP low,medium and high dose group and Benner Pury group at each time point in model group,were significantly increased?p<0.05?,compared with the model group,the kidney index of LBP low dose group and Benner Pury decreased significantly in the 7th?p<0.05?,the kidney index of LBP,high dose group and Benner Pury group decreased significantly in the 21th?p<0.05?;?2?the renal pathological change:compared with the sham operation group at each time point,the renal tubular interstitial injury index and collagen positive area was significantly increased?p<0.05?.Compared with UUO group,the tubulointerstitial injury index and the positive area of collagen staining in each LBP dose group and Benner Pury group decreased significantly?p<0.05?.?3?the results of immunohistochemistry:compared with the sham operation group,the levels of Wnt4,?-catenin,and TGF-?1 in the kidneys of other groups were significantly increased?p<0.05?;Compared with the model group,the expression of Wnt4,?-catenin and TGF-?1 in the LBP low,middle and high dose groups and Benner Pury group rats were significantly lower?p<0.05?;?4?Real Time PCR results:compared with the sham group,the mRNA expression of Wnt4,?-catenin and TGF-?1 in the kidneys of other groups was significantly increased?p<0.05?;Compared with the model group,,the expression of Wnt4,?-catenin,and TGF-?1 mRNA in the LBP low,middle,high dose groups and Benner Pury group were significantly lower?p<0.05?.Conclusion LBP can reduce the degree of tubulointerstitial injury in UUO rats,reduce the expression of Wnt4,?-catenin and TGF-?1 in the Wnt/?-catenin signaling pathway,reduce the inflammatory state of the kidney,and delay the progression of RIF.
