Objective:Using different doses of STZ respectively to make animal model of type 2 diabetes,the model success rate of different groups was compared to find out the optimal injection dose of STZ,and the effect of poria cocos complex extract on blood glucose,lipid level and antioxidant capacity of rats with type 2 diabetes mellitus was discussed based on this model,so as to elucidate its mechanism of action.Method:Type 2 diabetes animal model: Male SD rats of SPF grade of 4 weeks old were divided into:normal control group(10 rats),high-fat feeding group(70 rats),the normal control group was given maintenance feed,and the high-fat feeding group was given high-fat feed.After 4 weeks,the rats of the high-fat feeding group were randomly divided into four groups: high-fat control group: 50mg/kg citric acid buffer was injected intraperitoneally(10 rats);Model?groups: intraperitoneal injection of 30 mg/kg STZ(20 rats);Model ? groups: intraperitoneal injection of 40 mg/kg STZ(20 rats);Model ? groups: intraperitoneal injection of 55 mg/kg STZ(20 rats).The normal control group was injected with citric acid buffer solution 50mg/kg intraperitoneally.At the end of the experiment,various biochemical indexes of rats in each group were detected,and the model success rate of rats in each model group was compared to find out the optimal model dose.Effect of poria cocos complex extract on blood glucose and lipid levels in type 2 diabetic rats: 65 male SD rats of SPF grade of 4 weeks old only adaptive feed after 1 week,10 rats were randomly selected as blank control group,the remaining 55 were used for the preparation of type 2 diabetes model.55 rats were fed high-fat diet for 4 weeks,at the end of the 4th week,the rats fasted for 12 h and received a one-time intraperitoneal injection of 1% STZ at a dose of 30 mg/kg.After that,the high-sugar and high-fat feed was fed for another week,and the fasting blood glucose was detected.The model was successfully modeled based on the blood glucose value > 11.1 mmol/L.The rats were randomly divided into 5 groups:model control group,metformin group,poria cocos low dose group,poria cocos medium dose group,and poria cocos high dose group.All rats lavage 4 weeks in a row,detection of fasting blood glucose,triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),insulin(INS),glucagon(GC)levels,serum superoxide dismutase(SOD),malondialdehyde content(MDA),and the homeostasis model was calculated to evaluate the insulin resistance index(HOMA-IR),observation of the pancreas,liver tissue change form.Result:1.Different doses of STZ combined with high-fat feeding could successfully establish the rat model of type 2 diabetes.and the optimal dose of STZ is 30 mg/kg.2.Compared with the model control group,the weight of poria cocos complex in the low,medium and high dose groups was significantly increased(P <0.05),and the fasting blood glucose was significantly reduced(P <0.05).3.Serum TG,TC and LDL-C of the low,medium and high dose poria cocos complex group were decreased than that in model control group(P <0.05).HDL-C was higher than model control group(P < 0.05).4.Compared with the model control group,INS in the low,medium and high dose groups were significantly increased(P < 0.05),GC was significantly decreased(P < 0.05),and HOMA-IR index was significantly decreased(P < 0.05).5.Compared with the model control group,the activity of serum superoxide dismutase increased and the content of malondialdehyde decreased of the low,medium and high dose poria cocos complex group(P <0.05).6.HE staining results showed that the arrangement of islet cells and diffuse steatosis of liver cells in the low,medium and high dose gavage groups of poria cocos were improved to varying degrees.Conclusion:1.STZ 30 mg/kg was intraperitoneally injected with the highest mold formation rate.2.The extract of poria cocos can maintain diabetes rats normal body weight,reduce the fasting blood sugar,improve insulin resistance.3.The extract of poria cocos can regulate blood lipid of diabetic rats and improve the antioxidant capacity of the body.4.The extract of poria cocos can improve the islet function and liver injury in diabetic rats.
