Purpose: To investigate the effects of pectin and sucralfate on the expression of MTL and CGRP-mRNA in rats with gastric ulcer and the effects of gastric ulcer index and ulcer inhibition rate.Methods: Forty-eight SPF male Wistar rats were randomly divided into control group,model group,pectin group and sucralfate group,with 12 rats in each group.Rats in the model group,pectin group and sucralfate group were prepared by glacial acetic acid method.The rats in the blank control group were operated in the same procedure,but only the abdominal cavity was opened,and the sterilized water for injection was used instead of glacial acetic acid.After the second day of modeling,the rats were intragastrically administered.The rats in the pectin group were given pectin by gavage,the rats in the sucralfate group were given sucralfate by gavage,and the rats in the blank group and the model group were given normal saline by gavage.1 time / day,continuous gavage for 14 days.After the last intragastric administration,the rats were fasted for 24 hours,anesthetized and sacrificed,the abdominal cavity was cut,the stomach tissue was dissected,and the sterile saline was rinsed off.The 10% formaldehyde solution was fixed,and the ulcer index and ulcer were measured and calculated.Index and ulcer inhibition rate.The gastric mucosa of the ulcer was harvested and the tissue homogenate was prepared.The expression levels of MTL in the tissues were measured by kit.The expression levels of CGRP-mRNA were measured by RT-PCR.The pathological sections of HE staining were prepared for histopathological observation.Experimental data was processed using SPSS 19.0 statistical software.Result:(1)Compared with the blank control group,the MTL level in the gastric ulcer tissue of the model group was significantly increased(P<0.05).The CGRP-mRNA level in the gastric ulcer tissue of rats was significantly decreased(P<0.05).Rat gastric ulcer area increased significantly(P<0.05),ulcer index increased significantly(P<0.05);rat gastric ulcer was typical round or Oval shape,crater-like,gastric mucosa can be seen as a star-shaped shape of the ulcer,and the mucosal fold wall becomes thicker;the mucosal defect and the gland disappeared at the ulcer,and there are a large number of inflammatory cells and necrotic exudate at the bottom of the ulcer.Obvious granulation layer and scar tissue.(2)Compared with the model group,the MTL levels in the gastric ulcer tissue of the pectin group and the sucralfate group were significantly decreased(P<0.05).The MTL level in the gastric ulcer tissue of the pectin group was significantly lower than that in the sucralfate group(P<0.05).(3)Compared with the model group,the CGRP-mRNA levels in the gastric ulcer tissue of the pectin group and the sucralfate group were significantly increased(P<0.05).The CGRP-mRNA level in the gastric ulcer tissue of the pectin group was significantly higher than that in the sucralfate group(P<0.05).(4)Compared with the model group,the area of gastric ulcer in the pectin group and the sucralfate group was significantly decreased(P<0.05),and the ulcer index was significantly decreased(P<0.05).The area of gastric ulcer in the pectin group was significantly smaller than that in the model group.In the sucralfate group(P<0.05),the ulcer index was significantly lower than that in the sucralfate group(P<0.05),and the ulcer inhibition rate was significantly higher than that in the sucralfate group(P<0.05).(5)The gastric mucosal epithelial cells in the pectin group and the sucralfate group were significantly reduced,the gastric mucosal integrity was good,the ulcer was flat;the ulcer area was reduced,the marginal regenerating gland was partially cystic,and the surface of the ulcer was regenerated.The gland is covered,and the connective tissue between the glands is reduced;the infiltration of chronic inflammatory cells is reduced,and the new capillaries are abundant.The pathological grade of rats in the pectin sputum group was due to the sucralfate group.Conclusion: Pectin and sucralfate can effectively improve the MTL and CGRP-mRNA levels in rats with gastric ulcer,and effectively reduce the ulcer area and ulcer index of rats with gastric ulcer.The ulcer inhibition rate is higher than that of sucralfate.What,s more,Pectin can improve the pathology of ulcer gastric mucosa.
