| Objective:To investigate the effect of competitive inhibition ofβ-catenin/TCF andβ-catenin/FoxO1 signal downstream of transforming growth factor-β1 on angiogenesis and plaque stability in carotid plaque.Methods:1.Plaque tissue of patients with carotid stenosis after CEA(n=21).Vulnerable and stable plaques were identified by hematoxylin-eosin(HE),and the expression sites and levels of PECAM1/CD31 and VE-Cadherin in each tissue were analyzed by immunohistochemical(IHC).2、Model of carotid stenosis was established in ApoE-/-mice(Group A、B、C、D,n=12/group).Group A:TGF-β1(50ug/kg)+normal saline(5mg/kg);Group B:TGF-β1(50ug/kg)+ICG-001(5mg/kg);Group C:normal saline(50ug/kg)+ICG-001(5mg/kg));Group D:normal saline(5.05mg/kg).All group were treated by their own treatment for 2 weeks,then the carotid tissues were taken.After hematoxylin-eosin(HE),vulnerable and stable plaques were divided into vulnerable and stable plaques according to pathological characteristics.The expression sites and levels of PECAM-1/CD31 and VE-Cadherin in each tissue were stained by immunohistochemical(IHC).All data were analyzed by Imageplus6.0 and SPASS25.0 statistical software.Results:The content of TGF-βin stable plaque is higher than that in vulnerable plaque.But the expression of VE-Cadherin and CD31 in stable plaque are less than vulnerable plaque(p<0.05).In animal experiment,the rate of vulnerable plaque in group A(TGF-β)and group B(TGF-β+ICG-001)is lower than that in blank control group.And the positive expression of VE-Cadherin and CD31 in group B(TGF-β+ICG-001)are lower than that in group A(TGF-β)and blank control group(p<0.05).Conclusion:Specific blockingβ-catenin/TCF downstream of TGF-βcan inversely enhance the inhibitory effect ofβ-catenin/FoxO1 on carotid atherosclerosis intraplaque angiogenesis,thereby having a positive impact on stabilizing plaque. |
