Background and purposeDrug-Eluting Beads embolization technology is the forefront of current interventional research and is being widely used in the treatment of liver cancer.The most critical factor for the success of this technology is the efficacy of Drug-Eluting Beads.Curcumin-Loaded Microspheres is a new type of Drug-Eluting Beads product that has not yet been used in clinical applications.This study evaluated the inhibitory effect of Curcumin Drug-Eluting on vx2 rabbit liver tumors and its effect on the life cycle of tumor-bearing rabbits through animal experiments.Materials and Methods1.Establish an experimental rabbit liver cancer animal model.2.Curcumin-Loaded Microspheres were injected into the hepatoma's blood supply artery by DSA and Superselective Catheterization,observe and record the general situation,calculate the difference in the survival cycle between Curcumin-Loaded Microspheres embolization group and the control groups.3.Review CT plain scan and dual-phase enhanced scan 21 days after surgery,using the formula(V):V=1/2×ab~2(a represents the maximum diameter of the tumor,b represents the minimum diameter of the tumor)calculate the tumor volume,compare with Blank Microsphere embolization group and Angiography group,observe and analyze the effect of Curcumin-Loaded Microspheres on liver cancer.4.After the VX2 liver cancer rabbits of each group died,the liver was removed,and the gross observation was performed first,and then a professional technician in the pathology department performed HE staining to observe the microsphere deposition,the presence of necrotic tissue and the survival of tumor cells.Result1.The first postoperative deaths in Curcumin-Loaded Microspheres embolism group,Blank Microsphere embolism group,and Angiography group occurred on day 38,day 27,and day 26.The average survival time of each experimental group was Angiography group 32.40days,Blank Microsphere embolization group 37.60 days,Curcumin-Loaded Microspheres embolization group 47.80 days.The median survival was 31 days for Angiography group,39days for Blank Microsphere embolization group,46days for Curcumin-Loaded Microspheres embolism group.After a pair-wise comparison,it was concluded that Curcumin-Loaded Microspheres embolism group was better than Angiography group(P<0.05)and better than Blank Microsphere embolism group(P<0.05),the difference was statistically significant.Compared with Angiography group,Increase of Life Span(ILS)of the two embolization treatment groups was 46.80%in Curcumin-Loaded Microspheres embolization group and36.60%in Blank Microsphere embolization group.The ILS between the two groups was significantly higher in Curcumin-Loaded Microspheres embolism group than in Blank Microsphere embolism group(P<0.05).2.Comparing the tumor data of the three groups of experimental rabbits 21 days after interventional surgery and 7 days before the operation,the tumor lengths and short diameters of the three groups of experimental rabbits increased in varying degrees.The volume was calculated with reference to the calculation formula and compared with the analysis:The tumor volume growth rate of Curcumin-Loaded Microspheres embolization group was476.00%,the smallest value,and the tumor volume growth rate of Angiography group was1247.83%,the difference was statistically significant(P<0.05),the volume growth rate of Blank Microsphere embolism group was 803.03%,which was between other two groups.After a pairwise comparison,it was concluded that Curcumin-Loaded Microspheres embolization group was better than Angiography group(P<0.05)and better than Blank Microsphere embolization group(P<0.05),and Blank Microsphere group was better than Angiography group(P<0.05),the difference was statistically significant.3.Under Optical Microscope,in Angiography group,a large number of cancer tissues were diffusely distributed.The cancer cells were arranged in adenoid-like solids,the cancer cells were polygonal,the cytoplasm was rich,granular,eosinophilic,the nuclear-plasmic ratio was large with hyperchromatic nuclei.In Blank Microsphere embolization group,scattered embolic microspheres were seen in the blood vessels,and the shape was regular without deformation.The surrounding cancer tissue was diffusely distributed,with less interstitial,and the liver cancer cells were arranged in adenoid-like solid form,with a small amount of necrosis.In Curcumin-Loaded Microspheres embolization group,microsphere deposition was visible in the blood vessels,and the morphological results were similar to those of Blank Microsphere embolization group.The surrounding cancer tissues were reduced to varying degrees,replaced by fibrosis,and necrosis was seen.Conclusion1.After embolization with Curcumin-Loaded Microspheres,tumor growth can be effectively inhibited by blocking the blood supply to the tumor tissue and the slow-release effect of curcumin.2.Curcumin-Loaded Microspheres embolization can cause cancer cells to show different degrees of necrosis and fibrosis,and achieve the purpose of reducing cancer tissue.3.The experimental rabbits in Curcumin-Loaded Microspheres embolization group recovered well after surgery without related complications.4.Curcumin-Loaded Microspheres embolization can significantly improve the survival cycle of liver cancer rabbits.
