Background HCC is a kind of primary liver cancer with high mortality.In China,cancer mortality ranks the third.It is known for its high incidence rate and high mortality worldwide.The formation of HCC is characterized by many factors,stages and synergistic effects.At present,the pathogenic factors and pathogenesis of HCC have not been fully studied.Therefore,a thorough study of the specific molecular mechanism of HCC is of great significance for early prevention,early diagnosis and accurate treatment of HCC.More and more LncRNAs have been confirmed to play a regulatory role in the formation of HCC through MAPK pathway.Therefore,it is of great significance to explore the carcinogenic mechanism of LncRNAs for the development of diagnostic markers and new therapeutic targets of HCC.Method(1)The expression of LncRNA LINC00978 in HCC and paracancerous tissues was detected by real-time fluorescent quantitative PCR(QRT-PCR).The correlation between the expression level of LncRNA LINC00978 and clinical data was demonstrated by statistical software.(2)The expression of LncRNA LINC00978 in normal hepatocytes and different hepatoma cell lines was detected by QRT-PCR.According to the experimental results,Hep3 B and Huh7 were selected to construct the knockdown cell model of LncRNA LINC00978.MTT assay,flow cytometry and colony forming assay were used to detect the cell model.To explore the effect of knockout of LncRNA LINC00978 on the proliferation,invasion,migration and cell cycle of Hep3 B and Huh7 HCC cells.The total protein of the model was extracted and Western blot was used to detect the expression of key molecules and their phosphorylation level in MAPK signal pathway after knockout of LncRNA LINC00978.Result(1)The expression of LncRNA LINC00978 in 49 clinical samples was detected by QRT-PCR.The expression level of LncRNA LINC00978 was increased in HCC and decreased in paracancerous liver.The expression of LncRNA LINC00978 in patients with advanced HCC was significantly higher than that in patients with early HCC.The overall 5-year survival rate of the high expression group was significantly lower than that of the low expression group.(2)The expression of LncRNA LINC00978 in HCC cells was significantly higher than that in normal hepatocytes.(3)The knockdown model of LncRNA LINC00978 mediated by shRNA showed that the ability of proliferation and invasion of cancer cells was significantly reduced after knockdown of LncRNA LINC00978;the apoptosis of cancer cells was significantly increased and the cell cycle was blocked after knockdown of LncRNA LINC00978.(4)Western blotting showed that knockdown of LncRNA LINC00978 decreased the transformation of ERK,p38 and JNK,the key proteins of MAPK signaling pathway,into phosphorylated p-ERK,p-p38 and p-JNK,and affected the activity of the pathway.Conclusion(1)LncRNA LINC00978 is a cancer promoting gene,which is highly expressed in HCC tissues and promotes the occurrence and development of HCC.The expression of LncRNA LINC00978 was positively correlated with the stage of the disease.High expression of LncRNA LINC00978 is associated with poor prognosis in HCC patients.(2)LncRNA LINC00978 regulates the phosphorylation of ERK,p38 and JNK,which are key molecules in MAPK signaling pathway,and then affects the occurrence and development of HCC.
