| Objective:To study the effect and mechanism of long non-coding RNAAC098934 on the invasiveness of non-small-cell lung cancer(NSCLC).Methods: The expression level of LncRNAAC098934 in either the NSCLC or the normal tissues was identified in the TCGA database.Transwell assay detected human non-small cell lung cancer(NSCLC)cell lines A549 and H1299 transfected with si RNA,and divided them into four groups,namely CON group,NC group,si-RNA1 group and si-RNA2 group,and detected their aggressiveness.LncRNAAC098934 was transfected into A549 and NCI-H1299 cells,and they were divided into 3 groups,namely NC group,OE-AC098934 group and OE+SB203580 group,and the changes in invasiveness were detected by Transwell assay.Western Blot was used to detect the expression levels of p38,p-p38,JNK,p-JNK,Myc and p53 proteins in A549 and NCI-H1299 cell samples inhibiting LncRNAAC098934.Results:Compared to the normal tissues,LncRNAAC098934 was more highly expressed in NSCLC tissues.Inhibiting the expression of LncRNAAC098934 could significantly inhibit the invasiveness of A549 and H1299 cells.After overexpression of LncRNAAC098934,the invasiveness of A549 and NCI-H1299 cells was significantly enhanced(P<0.05),while the invasiveness of A549 and NCI-H1299 cells was inhibited after inhibiting p38 MAPK pathway.In A549 and NCI-H1299 cells that inhibited the expression of LncRNAAC098934,the protein expressions of Myc,p-p38/p38 and p-JNK/JNK were significantly increased(P<0.05),and p53 protein was significantly decreased(P<0.05).Conclusion:LncRNAAC098934 enhances the invasiveness of NSCLC by activating the p38 MAPK pathway. |
