Objective Study on the characteristics of gut microbiota imbalance in PLGC model rats and explore the mechanism of PLGC on the imbalance of flora in the treatment of PLGC disease.Method 120 SD male rats were randomly divided into blank control group 20 and model control group 100.The model group was modeled by the MNNG composite modeling method.After the model was proved to be successful,it was randomly divided into treatment groups with different concentrations of compound lizard powder?0.07 g/ml,0.14g/ml,0.28g/ml?,vitamin control group,model group control group,and blank control group.Except the blank group,the other groups were continuously intervened for 12 weeks.The feces of rats in each group were collected aseptically in batches after successful modeling,before treatment?0 weeks of treatment?,during treatment?4 weeks of treatment?,after treatment?12 weeks of treatment?.HE pathological analysis of gastric mucosa tissue,and 16 S r RNA gene sequencing analysis of fecal samples.Result 1.Physiological and pathological manifestations of rats after treatment In the treatment groups of the compound lizard powder,the physiological and pathological conditions of the rats were significantly improved after the drug intervention,and the symptoms such as flatulence disappeared.Compared with the model group,the mucosal atrophy of some rats in each treatment group of the compound lizard powder was repaired,the intestinal metaplasia was improved,and the dysplasia was subsided.Among them,the compound lizard powder medium dose group?MD group?had a more significant effect.2.Differences in gut microbiota between PLGC model rats and blank rats induced by MNNG complex method?1?Changes in alpha diversity: Compared with the blank group,the Simpson index?p =0.0284018370442?showed that the colony diversity of the two groups was significantly different,and the model group had significantly lower colony richness than the blank group.?2?Beta diversity analysis: Firmicutes are the most abundant gates in BC and MG?86.37% ±4.14% vs.68.44% ± 23.89%?,followed by Bacteroidetes?11.57% ± 3.99% vs.21.57 ±16.88%?,at the gate level Firmicutes in the blank group were significantly higher than the model group,while Bacteroidetes were lower than the model group.At the genus level,Lactobacillus is most abundant in BC and MG?33.59% ± 14.78% vs.17.55% ± 11.24%?.Lactobacillus in the blank group is significantly higher than the model group,while Bacteroides is lower than the model group.At the gate level,Firmicutes enriched BC and Proteobacteria enriched MG.At the genus and species level,Lactobacillus,Christensenella and Ruminococcus bromii were enriched in BC,while Bilophila,Phascolarctobacterium,Prevotella and Ruminococcus flavefaciens were enriched in MG.3.Effects of different concentrations of compound lizard powder on gut microbiota in PLGC rats After treatment,7 bacterial genera were found to have changed significantly,namely Akkermansia,Bacteroides,Coprococcus,Lacobacillus,Prevotella,Ruminococcus,and Turicibacter.In each treatment group,the content of Akkermansia in the VC12W?P =0.0465?group decreased;Increased Bacteroides content in LD12W?P = 0.0214?;Coprococcus decreased at LD4W?P = 0.003?and LD12W?P = 0.0002?,but increased at HD4W?P = 0.0002?and HD12W?P = 0.0082?;Lactobacillus decreased in LD4W?P =0.0029?and increased in VC4W?P = 0.0007?,MD4W?P <0.0001?and MD12W?P<0.0001?;Prevotella decreased at MD4W?P = 0.0144?and MD12W?P <0.0001?,while at VC12W?P = 0.0029?,HD4W?P <0.0001?and HD12W?P <0.0001?increased;Ruminococcus decreased in VC4W?P = 0.029?,VC12W?P = 0.0005?,LD12W?P = 0.0005?,MD12W?P = 0.0014?,and HD12W?P = 0.0063?.Turicibacter decreased in HD4W?P <0.0001?and HD12W?P <0.0001?and increased in LD12W?P = 0.0041?.Conclusion 1.Compound Lizard powder can improve the overall physiological status of rats with gastric precancerous lesions,such as water intake,mental state and survival,and improve the pathological status of model rats,such as atrophy,intestinal metaplasia,and dysplastic gastric mucosal glands Physical condition.2.PLGC model rats have imbalance in the gut microbiota,mainly from normal consumption bacteria?Firmicutes?to more volatile bacteria?Proteobacteria?.3.The compound lizard powder gel can treat PLGC diseases through two-way regulation of gut microbiota,such as multiplying beneficial bacteria,inhibiting harmful bacteria,balancing gut microbiota structure.4.The Chinese herbal medicine compound may play a supporting role in probiotics by strengthening the spleen,which can promote the growth of intestinal probiotics?Akkermansia,Lactobacillus,Bacteroides?.The mechanism may be similar to that of supplementary Chinese medicines that contain polysaccharides and have probiotics.And which via detoxifying and detoxifying,reducing qi,promoting gastrointestinal peristalsis,reducing the residence time of harmful bacteria?Prevotella,Coprococcus,Ruminococcus,Turicibacter?in the intestine,and inhibiting the growth of pathogenic bacteria.
