This experiment uses precious Chinese medicine Dendrobium officinale as the main material,and uses intraperitoneal injection of chemical drugs(tetraoxopyrimidine and streptozotocin)to construct a mouse model of type 2 diabetes,and explores its hypoglycemic effect on type 2 diabetes mice.Dendrobium candidum as the main material to assist in the treatment of diabetes nutrition and health products.Dendrobium officinale polysaccharides(DOP)on the hypoglycemic effect of type 2 diabetic mice:(1)The fasting blood glucose value of model group mice was 48.4 ± 3.56(mmol / L)in the fourth week The fasting blood glucose values of the coarse and fine powder groups of mice were 12.93 ± 2.13(mmol / L)and 12.68 ± 2.31(mmol / L),indicating that DOP has a significant hypoglycemic effect on type 2diabetic mice(P <0.05).(2)At the end of the experiment,the insulin content and insulin resistance of the model group mice were 14.66 ± 2.64(m IU / L)and 18.32 ± 1.58,respectively,while the fine powder group was 17.13 ± 1.33(m IU / L)and 12.13 ± 2.35,indicating DOP can significantly increase the level of serum insulin in type 2 diabetic mice and reduce its insulin resistance(HMOA-IR).(3)Shannon and Simpson indexes of mice treated with DOP were 6.9366 and 0.9236,respectively,while Shannon and Simpson indexes of model mice were 5.9333 and0.9678,respectively.It shows that DOP can increase the diversity of intestinal flora in diabetic mice.In the model group,the abundance of Lactobacillaceae in the intestine of mice is about 5%.After DOP treatment,the abundance of Lactobacillaceae in the mouse intestine is about 30%;in addition,Akkermansia From no increase in the model group to about 4%,indicating that DOP can increase the abundance of beneficial bacteria in mice intestines..Dendrobium officinale compound(DOC)on hypoglycemic effects intype 2 diabetic mice:(1)At the end of the experiment,the fasting blood glucose of the model group mice was 27.11 ± 543(mmol / L),while DOC The fasting blood glucose value of mice in the high-dose group was13.12 ± 5.01(mmol / L),indicating that DOC has a significant hypoglycemic effect on type 2 diabetic mice(P <0.05).(2)At the end of the experiment,the insulin content and insulin resistance of the model group mice were 13.58 ± 2.46(m IU / L)and 18.85 ± 1.35,respectively,while those in the high-dose group were 21.49 ± 2.13(m IU / L)and 10.42± 2.35,indicating that DOC can significantly increase serum insulin levels and reduce insulin resistance(HMOA-IR)in type 2 diabetic mice.(3)Real-time quantitative PCR results in the liver of type 2 diabetic mice showed that compared with the normal group,the expression levels of Glut-2,Gck and Pklr in the model group were significantly down-regulated,respectively 0.42 ± 0.06 times of the normal group,0.31± 0.04 times and 0.28 ± 0.03 times;compared with the model group,the expression levels of Glut-2,Pklr and Gck in the liver of mice after DOC treatment were up-regulated,and the high-dose group of DOC reached0.81 in the normal group ± 0.06 times,0.83 ± 0.05 times,and 0.76 ± 0.11times;real-time quantitative PCR results in the pancreas of type 2diabetic mice showed that compared with the normal group,the expression levels of Pdx1 and Ins1 in type 2 diabetic mice were significantly down-regulated,They were 0.48 ± 0.04 times and 0.42 ±0.02 times of the normal group(P <0.01).After DOC treatment,the high-dose group of DOC reached 0.79 ± 0.11 times and 0.87 ± 0.15 times of the normal value(P <0.05).Compared with the normal group,the expression of Bcl2 in the pancreas tissue of the model group was significantly decreased,which was 0.60 ± 0.03 times of the normal group(P <0.05),while the expression level of the Caspase 3 gene in the model group was significantly increased,which was the normal group.1.42 ±0.11 times(P <0.05),the expression of this gene was down-regulated after DOC treatment,which was 1.11 ± 0.11 times of the normal group,no significant difference compared with the normal group.
