Objective: To explore TS mRNA expression in relation to prognosis of metastatic colorectal cancer patients received 5-FU based chemotherapy.Methods: A total of 66 patients with metastatic colorectal cancer in the Affiliated Drum Tower Hospital of Nanjing University Medical School from November 2013 to February 2018 were involved in this study.Patients' clinical pathological parameters,expression of TS mRNA,chemotherapy regiment were systematically.Univariate analysis and multivariate analysis were used for analyzing the relationship between TS mRNA expression and patients' clinical pathological parameters,independent prognostic factor of PFS and OS in metastatic colorectal cancer.Subsistence analysis was used for analyzing the relationship between TS mRNA expression and PFS/OS of patients with metastatic colorectal cancer.Results: 66 patients with metastatic colorectal cancer were involved in this study.Univariate analysis showed that vessel invasion(p(27)0.05),nerve invasion(p=0.001),abdominal cavity metastasis(p=0.026)were accosiated with TS mRNA expression.However,patients' age(p=0.251),gender(p=0.732),tumor location(p=0.758),T stage(p=0.912),differentiation(p=0.241),N stage(p=0.262)were not accosiated with TS mRNA expression.53 patients received XELOX regiment,13 patients received mFOLFOX regiment.The median overall survival of patients who received XELOX was 21 months,mFOLFOX was 24 months.The median progress-free survival of patients who received XELOX was 10 months,mFOLFOX was 10 months.There was no significant difference in OS and PFS between them(p=0.702,0.587,respectively).Multivariate analysis showed that age(p=0.019),TS mRNA expression(p=0.000),vessel invasion(p=0.002),T stage(p=0.001),liver metastasis(p=0.007),abdominal cavity metastasis(p=0.007)were the independent prognostic factors of OS,and tumor location(p=0.012),TS mRNA expression(p=0.001),T stage(p=0.003),liver metastasis(p=0.006),abdominal cavity metastasis(p=0.011),pelvic cavity metastasis(p=0.032)were the independent prognostic factors of PFS in patients with metastatic colorectal cancer.The median overall survival in high group of TS mRNA expression was 21 month,low group was 24 months.Patients with low expression of TS mRNA had longer OS than those with high expression of TS mRNA(p=0.006).The median progress-free survival in high group of TS mRNA expression was 8months,low group was 10 months.Patients with low expression of TS mRNA had longer PFS than those with high expression of TS mRNA(p=0.004).Conclusion: TS mRNA expression level may be a prognostic indicato in patients with metastatic colorectal cancer.Moreover,TS mRNA expression level might be a potential biomarker to predict the efficacy of 5-FU based chemotherapy regiments in patients with metastatic colorectal cancer.Patients with high TS mRNA expression had poor prognosis compared with those with low expression of TS mRNA.
