Objective:Through Morris water maze experiment,the behavioral changes of rats were studied to study the effect of parexibe sodium on cognitive function after operation,and the effect of parexibe sodium on stress response was studied by measuring plasma cortisol and blood glucose concentration of rats.Methods:Sixty healthy male SD rats were selected and divided into control group,sham operation group,preoperative group(preoperative administration group and preoperative water supply group),postoperative group(postoperative administration group and postoperative water supply group).All rats were tested with water maze to monitor the escape latency,target quadrant dwell time and the number of times of piercing.In addition,210 healthy male SD rats were divided into control group,sham operation group,preoperative group(preoperative administration group and preoperative water supply group),postoperative group(postoperative administration group and postoperative water supply group).In sham operation group,only bilateral common carotid arteries were separated,unclipped,and bilateral common carotid arteries were clamped before and after operation to prepare postoperative cognitive impairment(POCD)model.One week after feeding,the blood of abdominal aorta was extracted from the control group,and the other five groups were operated on one week after feeding.The blood of abdominal aorta was taken at 2 hours,1 day,3 days and 7 days after operation.The cortisol content was detected by ELISA Kit and the blood glucose concentration was detected by blood glucose meter.Results:The escape latency of rats in each group is shortened at each time point before operation,and there is no significant difference between the groups(P > 0.05).At different time points after operation,the escape latency of the control group was significantly lower than that of the preoperative and postoperative water supply groups(P < 0.05),with statistical significance.Compared with the preoperative administration group,the escape latency of the preoperative administration group was significantly shorter(P < 0.05),with statistical significance;compared with the postoperative administration group,the escape latency of the postoperative administration group was significantly shorter(P < 0.05),with statistical significance,and compared with the postoperative administration group,the escape latency of the preoperative administration group was significantly shorter(P < 0.05),with statistical significance.Compared with the control group,the rats in the water supply group and the water supply group had a significant reduction in the number of times of pierce and the time of stay in the target quadrant(P < 0.05).Compared with the postoperative group,the number of pierced rings and the staying time of the target quadrant in the preoperative group were significantly increased(P < 0.05).The changes of cortisol content and blood glucose in sham operation group,water supply group and drug administration group were basically the same,they all began to rise after operation,peaked at 2 hours after operation,and then decreased gradually.Compared with the sham operation group,the cortisol content and blood glucose of the rats in the water supply group were significantly higher than those in the sham operation group(P < 0.05).Compared with the water group,the cortisol content and the change trend and range of blood glucose in the two,one,three and seven days after operation in the drug group were significantly lower(P < 0.05).Compared with the postoperative administration group,the cortisol content and blood glucose in the preoperative administration group were lower than those in the postoperative administration group(P< 0.05).Conclusion:Parecoxib sodium can improve postoperative cognitive dysfunction by inhibiting the increase of cortisol and blood glucose concentration caused by stress response,and its preemptive analgesic effect is better than that of postoperative administration,which has guiding significance for its clinical application.
