Objective:To evaluate the caries activity of preschool children with ?-thalassemia major by using methods such as Streptococcus mutans 16 S rDNA PCR and Cariostat caries susceptibility test,and explore whether there is a correlation between preschool children with ?-thalassemia major and caries.To provide theoretical basis for the prevention and treatment of dental caries in preschool children with ?-thalassemia major,so as to implement targeted preventive health care measures and improve the quality of life of special population.Methods:A random sampling method was used to conduct questionnaire surveys and basic oral examinations of 32 children aged 3 to 6 years old with ?-TM who were in the Department of Pediatrics of Affiliated Hospital of Zunyi Medical University and 30 healthy children of matching age.The plaques were collected,and the acid producing ability of cariogenic bacteria in dental plaque was measured by Cariostat method,and the caries value was recorded.At the same time,saliva samples were collected,the pH value of saliva samples was measured with a pH meter,standard strains of Streptococcus mutans were cultured,bacterial group DNA was extracted from the standard strains and saliva samples,specific primers were designed for 16 S rDNA of Streptococcus mutans,and saliva samples were detected by PCR Streptococcus mutans,calculate the detection rate of S.mutans in saliva samples.Compared the dmft,CSI,Cariostat value,saliva pH value,and the detection rate of Streptococcus mutans between the 3-6 years old ?-TM group and the healthy group.The experimental data were collated and analyzed.Results:1.There was no significant difference in gender,age,eating habits before bedtime,number of teeth brushing and other basic data between ?-TM group and healthy control group(P>0.05).2.The number of children with ?-TM was higher than that of healthy children in the medium and severe danger zone.The difference between the two groups was statistically significant(P<0.05).The mean value of caries in the ?-TM group was higher than that in the healthy control group(P<0.05),and the difference was statistically significant.3.The average dmft index of the ?-TM group was higher than that of the healthy control group,and the difference was statistically significant(P<0.05).4.The CSI index of the ?-TM group was higher than that of the healthy control group,and there was no significant difference between the two groups(P>0.05).5.The pH value of saliva in the oral cavity of ?-TM children was lower than that in normal children,and the difference was statistically significant(P<0.05).6.The detection rate of Streptococcus mutans in the ?-TM group was higher than that in the healthy control group,and the difference was statistically significant(P<0.05).7.The prevalence of caries in the ?-TM group was higher than that in the healthy group,and the difference was statistically significant(P<0.05).The number of column connections was r = 0.273,but the correlation was weak.Conclusion:Preschool children with ?-TM have a correlation with caries,but the correlation is weak.
