Correlation Between TGF-?2 And Filtration Formation After Han And Kazakh Angle-closure Glaucoma

Object: To evaluate the expression levels of transforming growth factor ?2(TGF-?2)and the morphology of filtered vesicles in the aqueous humor and trabecular meshwork of patients with primary angle-closure glaucoma(PACG)of Han and Kazakh nationalities.Methods: Prospective clinical case-control analysis.A total of 46 49 eyes of Han and Kazakh PACG patients diagnosed in the Department of Ophthalmology of the First Affiliated Hospital of Shihezi University School of Medicine were included in 2018-07/2019-04,aged 40-85 years old,of which 25 were26 eyes of the Han nationality;21 were 23 eyes Eyes,there is no significant difference in age and gender between patients of the two nationalities.Aqueous humor and trabecular meshwork tissue samples were collected during trabeculectomy,and two methods were collected by immunohistochemistry(IHC)and immunofluorescence(IF).The content of TGF-?2 expression in the specimens was measured by ELISA method to detect the content of total aqueous TGF-?2(t TGF-?2)and activated TGF-?2(a TGF-?2).Changes in blood pressure,morphology of filtration vesicles,patient's age and t TGF-?2 level in aqueous humor after 6 months of follow-up.SPSS24.0 statistical software package was used for statistical data analysis,and P<0.05 was considered statistically significant.Results:There was no statistically significant difference in age,gender,postoperative type ? and ? type filter blebs between the two nationalities(P>0.05),and postoperative type ? and ? type filter bleb was statistically significant(P<0.05);70 years old The content of TGF-?2 in the aqueous humor of the above Han patients was significantly higher than that of the Kazakhs(P=0.026);the intraocular pressures were(13.91±2.95)mm Hg and(12.17±2.50)mm Hg in 3 months and 6 months after surgery.There are statistically significant differences in intraocular pressure between two kinds of local intraocular pressure-lowering drugs before and after operation(P<0.05,P<0.01);ELISA test results show that the difference of a TGF-?2 in the aqueous humor of PACG patients of Han and Kazakh nationalities is statistically significant(P <0.05);IHC and IF test results showed that the number of TGF-?2 positive cells in trabecular tissue cells of Han patients increased significantly,showing significantly high expression;using image software to perform cumulative fluorescence analysis of immunofluorescence of the two ethnic groups was statistically significant(P=0.018),which shows that TGF-?2 plays an important role in filtering scar formation.Conclusion:The expression of TGF-?2 in the aqueous humor and trabecular tissues of patients with PACG in the two ethnic groups is different,The expression of TGF-?2 in the aqueous humor and trabecular tissue of Kazakh patients is significantly lower,which reduces the promotion of postoperative filtering scar Functional filtration filtration bleb.
