Objective To investigate the changes of matrix metalloproteinase2(MMP-2) andthe inhibition of matrix metalloproteinase1(TIMP-1) in aqueous humor with aqueoushumor of primary angle closure glaucoma and calculate the ratio between MMPs andTIMPs, further to explain pathogenesis relationship between the dynamic balance andprimary closed angle glaucoma. Method The subjects were divided into experimental group and control group, theexperimental group were a total of15eyes of15patients with acute primary angleclosure glaucoma (group A), chronic primary angle closure glaucoma (B group) and12eyes of12patients and age-related cataract, as control group (group C) were16eyes of16patients. All patients were diagnosed definitely in the First Affiliated provinceHospital of Medical University of Anhui. The samples of aqueous humor were carriedout in operation by strict procedures and methods for collecting them.And the bloodwas collected in operation before1days, in15minutes to the centrifugal, getting thesupernatant and placing in the refrigerator to be detected at-80℃, using doubleantibody sandwich enzyme linked immunosorbent assay (enzyme linkedimmunosorbant assay, ELISA MMP-2) to determine the concentration of MMP-2and TIMP-1in aqueous humor and in the plasma,, and calculate ratio all the samplesbetween TIMP-1and MMP-2. Data were analyzed using SPSS10.0, by a=0.05for thetest, compared to MMP-2, TIMP-1concentration and ratio between TIMP-1andMMP-2. Whether the data difference has statistics significance.Results1, To express the data in mean standard deviation which was consistentwith normal distribution.In A, B, C group, the concentration of MMP-2in aqueous humor were (20.3291.827)(18.0012.073),(18.5301.631) ng/ml;In A, B, C group, the concentration of MMP-2in plasma were (4.8661.845)(5.3111.378),(4.7301.309) ng/ml;In A, B, C group, the concentration of TIMP-1in aqueous humor were (12.6523.018)(8.2831.721)(12.5794.013) ng/ml;In A, B, C group, the concentration of TIMP-1in plasma were (0.8220.564)(0.9280.400),(0.8430.630) ng/ml;In A, B, C group the ratios of TIMP-1and MMP-2in aqueous humor were (0.6290.172)(0.4690.128),(0.8870.164); In A, B, C group the ratios of TIMP-1and MMP-2in plasma were (0.2020.176)(0.1790.095),(0.1870.167);In A, B, C group, the ratio of MMP-2between plasma and aqueous humor were0.2360.078,0.3010.090,0.2590.080;In A, B, C group, the ratio of TIMP-1between plasma and aqueous humor were0.1000.130,0.1130.053,0.0550.037;2.Comparison of the indexes mentioned above, the TIMP-1/MMP-2in aqueous humorof PACG group has significantly difference compared to the control group (P <0.01),which is the ratio of TIMP-1/MMP-2decreased slightly compared to the age-relatedcataract in aqueous humor of PACG, the active MMPs in aqueous humor of PACGincreased, at the same time, suggesting that balance of MMPs and TIMPs is destroyed,showing that the molecular system in glaucoma may play a role in the development ofpathological process.3, The TIMP-1in aqueous humor of PACG were improved significantly compared tocataract, with statistical significance (P <0.01), namely, TIMP-1decreased comparedto the control group suggesting that Enzyme of TIMPs in PACG in the aqueous humoractivity effectively change.4, Other parameters compared with the control group had no statistical significance (P>0.05).Conclusions The systems of TIMP-1/MMP-2in aqueous humor of PACG aredamaged, suggesting that the MMP/TIMP system may play a role in the disease.
