Object: This study aimed to investigate the correlation between the expression of IL-6,STAT3,E-cadherin and N-cadherin proteins and Knosp classification in nonfunctioning pituitary adenoma(NFPA),in order to explore the relationship between these proteins expression and the invasiveness of nonfunctioning pituitary adenomas.Methods: 30 cases of pituitary adenoma patients admitted to the neurosurgery department of the first affiliated hospital of shihezi university medical college from 2013 to 2018 were collected.All specimens were confirmed as non-functional pituitary adenomas by Clinical manifestations,laboratory tests and pathological examination.According to the diagnostic criteria for the invasiveness of pituitary adenomas,20 cases of invasive nonfunctioning pituitary adenomas(INFPA)and 10 cases of noninvasive nonfunctioning pituitary adenomas(NNFPA)were included.Envision immunohistochemistry was used to detect the expression of IL-6,STAT3,E-cadherin and N-cadherin proteins in the specimens.SPSS 20.0 statistical software was used to evaluate the relationship between the above proteins and the invasiveness of nonfunctional pituitary adenoma.Results: Statistical analysis showed that the expression of IL-6,STAT3,E-cadherin and N-cadherin in non-functioning pituitary adenomas was not associated with gender and age.Immuohistochemical staining showed that IL-6 staining was localized to the cytoplasm which was brownish yellow and mainly diffuse existence.IL-6 expression in INFPAs was significantly higher than NNFPAs,the difference was statistically significant.STAT3 staining was mainly located in the cytoplasm and a small amount was localized in the nucleus.It was brown and diffuse.The expression of STAT3 in INFPAs was significantly higher than that in NNFPAs.The difference was statistically significant.E-cadherin staining was localized to the cytoplasm which was brownish yellow and mainly diffuse existence.The expression of E-cadherin in INFPAs was lower than that in NNFPAs.The difference was statistically significant.N-cadherin staining was mainly located in the cell membrane and a small amount in the cytoplasm which was mostly brown and diffuse.N-cadherin expression in INFPA was higher than NNFPA.The difference was not statistically significant.The expression of IL-6 and STAT3 was positively correlated with Knosp classification of pituitary adenoma;the expression of E-cadherin was negative with Knosp classification of pituitary adenoma.There was no significant correlation between N-cadherin expression and Knosp classification of pituitary adenoma.Conclusions: The IL-6 /STAT3 signaling pathway is related to the invasiveness of non-functional pituitary adenoma and its related proteins may become a potential biological marker to predict the invasiveness of pituitary adenoma,providing research directions for the future treatment of invasive pituitary adenoma.
