Objcetive:To explore the molecular pharmacological mechanism of salidroside in protecting rat heart injury caused by high altitude hypoxia.Methods:In this study,40 adult Wistar male rats were randomly divided into a blank control group?lanzhou,1500m height above sea level?and an anoxic group?6500m height above sea level?according to the random number table method.The anoxic group was further divided into a normal saline group,a salidroside group,a Compound C group,and a Compound C+salidroside grouph.According to the experimental design,the rats were weighed and the blank control group was raised in the animal experimental the 940 Hospital of joint Logistics support force of Chinese people's Liberation Army.Compared with the blank control group,each group of hypoxic rats had different degrees of myocardial damage,myocardial cells and myocardial interstitial edema,structural disorders and unclear cell boundaries.After 10 days,the number of deaths of the mice was counted,and the body weight was measured.After anesthetization,the rats were exposed to the abdominal aorta to draw arterial blood for blood gas index detection;further dissect and isolate the heart,and after HE staining,observe the changes of cardiomyocytes and myocardial fibers with a microscope;Terminal dexynucleotidy transferase?TdT?-mediated dUTP nick end labeling?TUNEL?Detection of cardiomyocyte apoptosis;Western blot detection of AMPK,P-AMPK,mTOR,P-mTOR and Beclin-1 protein expression of myocardial autophagy related pathways.Results:1.The body weight of the rats in the saline group,compound C group and salidroside+compound C group exposed to hypoxia was lower than that before hypoxia.Among them,the weight loss of anoxia saline group and compound C group was statistically significant?P<0.05,P<0.01?,but the weight of salidroside group increased,there was no significant difference in weight loss between salidroside+compound C group and that before hypoxia?P>0.05?.2.The levels of blood gas indexes pH,PaO2 and SaO2 of rats in each group exposed to hypoxic environment were lower than those in the normoxic group?P<0.01?;while the levels of PaCO2,Hb,HCT and Lac all increased compared with the normoxic group high?P<0.01?;the indicators of Compound C group changed more significantly,but the indicators of both groups improved after salidroside administration.3.The myocardial tissue of each group exposed to hypoxia also had different degrees of damage,myocardial cells and myocardial interstitial edema,structural disorder,unclear cell boundaries,at the same time,the number of myocardial cell apoptosis in each group was consistent with the degree of myocardial pathological damage.However,After salidroside treatment,the pathological morphology and the degree of myocardial cell apoptosis were improved.4.The expression level of AMPK and P-AMPK increased,while the expression level of mTOR and P-mTOR decreased.After administration of salidroside,the expression of AMPK and P-AMPK was further increased,and the expression of mTOR and p-mTOR was further decreased;The protein expression of compound C group was contrary to salidroside group;At the same time,the increase of AMPK and P-AMPK expression and the decrease of mTOR and P-mTOR expression in compound C and salidroside group were between compound C and salidroside group,but the expression of Beclin-1 was significantly higher than that of compound C?P<0.05?.Conclusion:1.Under the condition of simulated high altitude hypoxia,rats'weight loss,myocardial cell pathological damage and myocardial cell apoptosis increased.2.Salidroside can positively regulate AMPK/mTOR signaling pathway,activate autophagy,and play a protective role in anoxic rats.
