Objective:We investigate the effects of Inonotus obliquus aqueous extract on AMPK path in rats with type2diabetes. The type2diabetes is induced by high sugar and fat with a small dose of streptozotocin (STZ). We discuss the mechanism and effects of the Inonotus obliquus against type2diabetes. Our research improves the investigation on how Inonotus obliquus decreasing blood sugar, and provides experimental basis of healing type2diabetes for clinical applications.Methods:We select healthy mature male Winstar rats for experiments. We select ten rats as the normal control group randomly and others as experimental groups. After feeding rats in experimental groups fodder which is rich of high sugar and fat for four weeks, we give the rats single intraperitoneal injection of a small dose of STZ (25mg/kg) to induce type2diabetes. After one week, fasting but not for water. After twelve hours, we measure the fasting blood glucose (FBG) level. If FBG≥7.8mmol/L, the rat is confirmed as type2diabetes. The rats with type2diabetes are divided into five groups randomly. Respectively, these five groups are: group with type2diabetes (negative control group), positive control group, high concentration of Inonotus obliquus aqueous extract group, moderate concentration of Inonotus obliquus aqueous extract group, low concentration of Inonotus. obliquus aqueous extract group. We give metformin hydrochloride in positive control group and the concentration is200mg/kg. The rats in groups of different concentrations are given Inonotus obliquus aqueous extract(The concentrations are900mg/kg,600mg/kg,300mg/kg respectively.) by using stomach tube once a day for eight weeks. The rats in normal control group and the group with type2diabetes are given the same volume of water by using stomach tube once a day for eight weeks. At the end of the research, we measure the fasting blood glucose, glycosylated hemoglobin, glucose tolerance, AMPK activity in hepatic tissue and activities of PEPCK and G-6-PASE again.Results:In the results, Inonotus obliquus aqueous extract can decrease the blood sugar of the rats with type2diabetes which is induced by high fat and sugar with a small dose of STZ. And Inonotus obliquus aqueous extract can increase the activity of AMPK. We discover that the activities of PEPCK and G-6-PASE are related with the concentration of Inonotus obliquus.Conclusion:We induce type2diabetes on healthy mature male rats by high fat and sugar with the help of a small dose of STZ. We discover that Inonotus obliquus aqueous extract has outstanding effects against type2diabetes. We think it can activate AMPK and then decrease the activities of PEPCK and G-6-PASE in the path. In this case, gluconeogenesis is affected by Inonotus obliquus.
