Objective:A search of all relevant literature revealed that IL-22 plays an important role in heart disease.This experiment primarily discusses: 1.The change of IL-22 in ACS patients;2.The change of IL-22 in ACS patients according to the syntax score.Methods:Patients in the department of cardiology of jiujiang the 1st people's hospital were selected,and all the selected cases were divided into 20 cases in the Acute coronary syndrome(ACS)group and 20 cases in the stable angina pectoris(SAP)group according to the disease type.At the same time,20 healthy physical examinees(health control(HC)confirmed by electrocardiogram,myocardial marker and coronary angiography without coronary artery lesions were selected as the control group.Patients in the ACS group were divided into the low-risk group(score ?22),the medium-risk group(score > 22 < 32),and the high-risk group(score ?32)according to SYNTAX score,with 20 cases in each of the three groups.Real-time Flurescence Quantitative Reverse Transcription Polymerase Chain Reaction(RT-PCR)and enzyme-linked immunoassay(ELISA)were used to detect the levels of il-mrna and serum IL-22 in peripheral blood.Statistical software SPSS20.0 was used to process the experimental data,and P<0.05 or P<0.01 had statistical significance.Results:1.Compared with HC group,the serum IL-22 levels in ACS group and SAP group were increased,which were 31.41±7.06,17.10±4.81,and 9.38±2.45,respectively(P<0.01).The level of IL-22 in the ACS group was significantly higher than that in the SAP group(P<0.01).The levels of IL-22 mrna in peripheral blood PBMC in the ACS group and SAP group were higher than those in the HC group,which were 4.39±1.79,2.39±1.10,and 0.89±0.12,respectively(P<0.01).The level of IL-22 mrna in the ACS group was higher than that in the SAP group,and the difference was statistically significant(P<0.05).Correlation analysis showed that serum IL-22 had a significant positive correlation with IL-22 mrna in peripheral blood PBMC(r=0.70,P<0.0001).2.SYNTAX score: IL-22 mrna levels in serum IL-22 and peripheral blood PBMC in high-risk group were 41.33±6.57 and 6.58±1.01,respectively,which were higher than those in low-risk group,with statistically significant differences(P<0.01).The levels of IL-22 mrna in serum and peripheral blood PBMC in the high-risk group were higher than those in the medium-risk group,with statistically significant differences(P<0.01).The levels of IL-22 mrna in serum IL-22 and peripheral blood PBMC in the medium-risk group were 31.27±5.48 and 4.64±1.12,respectively,higher than those in the low-risk group(P<0.05).The differences were 23.10±6.25 and 3.09±0.85,respectively,in the low-risk group.Correlation analysis showed that serum IL-22 had a significant positive correlation with IL-22 mrna in peripheral blood PBMC(r=0.65,P<0.0001).3.The correlation analysis of IL-22 mrna in serum and PBMC in peripheral blood of low,medium and high risk patients with SYNTAX score group showed that SYNTAX score was significantly positively correlated with serum IL-22(r=0.74,P<0.0001),and SYNTAX score was also significantly positively correlated with serum IL-22 mrna in peripheral blood of PBMC(r=0.75,P<0.0001).Conclusion:The level of IL-22 was positively correlated with the severity of the disease in patients with acute coronary syndrome.
