Objective: To study the secondary metabolites of Penicillium sp.X R046 and their biological activities.Methods:(1)Penicillium sp.XR046 was cultured on rice medium,and the metabolites was extracted by ethyl acetate;(2)The compounds from metabolites of Penicillium sp.XR046 were isolated b y silica gel column chromatography(CC),Sephadex LH-20 gel CC,reversed-p hase CC,and preparative HPLC,and identified by spectroscopy techniques suc h as NMR,MS and CD;(3)The 96-well plate double dilution method was us ed to test the antibacterial activities of the compounds on standard and clinicall y resistant strains of bacteria.Results: A total of 30 compounds were isolated from the metabolites of Penicillium sp.XR046.of which 14 compouds named 3R-8-methoxy-3-(4-oxopentyl)isochroman-1-one(1),penicimarin B(2),penicimari n C(6),3R-7-hydroxy-8-methoxy-3-(4-oxopentyl)isochroman-1-one(7),3-hydrox ymethyl-6,8-dimethoxycoumarin(8),3R-(4S-hydroxypentyl)-5,6-dihydroxy-isochr oman-1-one(9),3R-(7,8-dihydroxy-1-oxoisochroman-3-yl)propanoic acid(10),6,8-dimethoxy-3-(2?-oxo-propyl)-coumarin(15),peniciisocoumarin H(19),penicill oxalone B(20),aspergillumarin B(21),pestalasin A(22),8-hydroxy-6-methoxy-3-methylisocoumarin(23)and aspergillumarin A(26)were isocoumarin derivati ves.Compounds 1 and 7 were the new isocoumarin compounds.Compounds o f berkeleytrione(13),chrodrimanin B(14),berkeleyacetal B(16),berkeleyaceta l C(18)and chrodrimanin E(30)were polyketone-terpenoid derivatives;cis-(3RS,4RS)-3,4-dihydro-2,4,8-trihydroxynaphthalen-1(2H)-one(11),3.4-dihydro-3.4.8-trihydroxy-1(2H)-naphthalennone(12)and regiolone(27)were tetralin derivativ es;12S,13R-Dihydroxy-9-oxo-octadeca-10(E)-enoic acid(3),(9E,11Z)14-hydroxyo ctadecan-9,11-dienoic acid(28)and nafuredin(29)are fatty acids.cyclo(D)-Pro-9,10-en-Phe(4)and cyclo(D)-Pro-8-N-methyl-9,10-en-Phe(17)are diketopiperazines.The remained compounds are 4-methyl-5,6-dihydropyren-2-one(5),O-meth oxyphenol(24)and 4-amino-2-methoxyphenyl acetate(25).Compounds 3?5?14?15?17?21?23?25?27?28 and 30 were isolated from the fungi Penicillium sp.for the first time.Compounds 1,2,6?10 were evaluated for antibacterial a ctivity against Candida albicans ATCC 5314 and Escherichia coli ATCC 25922 with MICs ranged from 50 to 100 ?g/m L;Compound 3 possessed antibacteria l activities against Escherichia coli ATCC 25922,Staphylococcus aureus ATCC25923,Staphylococcus epidermidis,and clinically isolated Escherichia coli and Staphylococcus aureus with MICs of 16?32 ?g/m L.Conclusion: Thirty compou nds were isolated from the secondary metabolites of Penicillium sp.XR046,tw o of which were new compounds.Some of the compounds are of potential val ue in the development of antimicrobial agents and provide experimental basis f or the search for biogenic antibiotics in coal area environment.
