Objective: To establish methods to distinguish ophiopogonis radix produced from two places and its adulterants by systematically pharmacognosy research on ophiopogonis radix and its adulterants.Methods: The pharmacognosy methods were used to analyze the original plant morphological characteristics of flowers,seeds,leaves,stems and roots,and the characteristics of the herbal medicine such as shape,color,texture and odor,cross-section and powder microscopic characteristics of different origins of ophiopogonis radix and its adulterants.Thin-layer chromatography was used to identify ophiopogonis radix and its confusing products(Fujian Mondo,Hubei Liriope,Broadleaf Liriope and Variegata).Standards were used to identify the chromatographic peaks of the fingerprints of Ophiopogon japonicus and its adulterants to establish ophiopogonis radix from Sichuan and Zhejiang,Fujian Mondo,Hubei Liriope,Broadleaf Liriope and Variegata HPLC fingerprints The results of thin-layer chromatography of Sichuan and Zhejiang ophiopogonis radix,Fujian Mondo,Hubei Liriope,Broadleaf Liriope and Variegata were studied by Chempattern and SIMCA software.Chemometric analysis was used to find differential components.Results: By observing the morphological characteristics of the original plants,a table of original plant morphological characteristics of four adulterants of different origins of ophiopogonis radix was established.The traits of 6 kinds of pieces were compared and established.By observing the cross-section of the roots of the different origins of ophiopogonis radix and its four adulterants and their microscopic characteristics under normal light and partial light,the microscopic feature retrieval tables of six kinds of decoction pieces were established.A thin-layer chromatography method was established based on the Hong Kong standards,and the method was examined Methodology research.Using thin-layer scanning technology to convert thin-layer spot chromatograms into chromatographic peaks,combined with stoichiometry to identify different sources of ophiopogonis radix and its adulterants.The chemical structure of the six common peaks was determined by summarizing the cleavage rules of the standards and comparing with the components contained in the samples.Using the newly established fingerprint method,the similarity between different batches of ophiopogonis radix or its adulterants is higher,but the similarity between themselves is low.Conclusions: The specific characteristics of plant morphology,character and microscopic structure of ophiopogonis radix and its adulterants were determined.The thin-layer chromatography and HPLC fingerprint identification methods of ophiopogonis radix and its adulterants were established.Provide reference for the authenticity identification,quality standard formulation,clinical application of Ophiopogon japonicus.
