Ophiopogon Steroidal Saponins Chemical Composition

We investigated constituent of steroidal saponins of the Ophiopogon japonicus (Thunb.)Ker-Gawl,one new compound together with four known compounds were isolated by usingopen-column(silica gel, macroreticular resin, ODS), preparative HPLC methods et al. The structure ofthe new steroidal saponin was elucidated as (25R)-spirostane-5-ene-3β, 14α-diol-3-O-α-L-xylopyranosyl (1â†'3) [β-D-glucopyranosyl (1â†'4)]-α-L-rhamnopyranosyl (â… ) by¹H-NMR,¹³C-NMR,HMBC,HMQC,¹H-¹H COSY,NOESY techniques, ESI/MS anzlysis,aswell as chemical methods. The structures of four known compounds were ophiogenin 3-O-α-L-rhamnopyranosyl (1â†'2)-β-D-glucopyranoside (â…¡), (25R) ruscogenin 1-O-[α-L-rhamnopyranosyl(1â†'2)] [β-D-xylopyranosyl(1â†'3)]-β-D-fucopyranosyl (â…¢,Ophiopogonin D), ophiogenin 3-O-α-L-rhamnopyranosyl (1â†'2) [β-D-xylopyranosyl (1â†'3)]-β-D-glucopyranoside(â…£), 24-O-β-D-fucopyranosyl-(23S,24S,25S)-23,24-dihydroxylruscogenin 1-O-[α-L-rhamnopyranosyl (1â†'2)][β-D-xylopyranosyl (1â†'3)]-α-L-arabinopyranoside(â…¥), by ¹H-NMR,¹³C-NMR, IR techniques.
