Objective: To observe the effects of Jianpigubenhewei Decoction on interstitial cells of cajal,c-kit and stem cell factor in gastric antral tissue of diabetic gastroparesis rats.To explore the effect and possible mechanism of Jianpigubenhewei Decoction in the treatment of diabetic gastroparesis.Methods: Sixty healthy male SD rats of SPF grade were randomly separated into normal group(10)and model group(50).The diabetic gastroparesis model was induced by onetime intraperitoneal injection of streptozotocin(STZ)combined with irregular feeding of high-fat and high-sugar diet in rats.When the blood glucose level was higher than 16.7 mmol / L,accompanied by anorexia,stool trait changes and weight loss,the gastric residual rate was significantly higher than that in the normal group to judge the success of replication of diabetic gastroparesis model.The successful rats were given intragastric administration according to different groups.The normal group and model group were given saline.The traditional Chinese medicine group was given high,medium and low doses of Jianpigubenhewei Decoction according to differrent groups,and the western medicine group was given domperidone suspension was administered continuously for 6 weeks.After 6 weeks,the gastric residual rate was detected.The distribution and expression of interstitial cells of Cajal,the protein expression levels of c-kit and SCF in gastric antral tissue were detected by immunohistochemistry and Western blot method.The contents of motilin,gastrin and Ghrelin in serum were measured by ELISA method.Result: 1.General situation: The normal group rats were in good condition,normal activity,lustrous fur,normal food intake and drinking water,no significant changes in urine and fecal properties,and increased body weight.DGP model group rats appeared to drink more,eat more,urine and defecate more significantly,weight gain slowly or lighter than before,and their fur was not lustrous,they were low in spirits and act slowly.At the later stage of the model,rats in each group began to eat less,stool appeared be irregular.After two weeks of drug intervention,rats in the high,medium and low dose groups of Jianpigubenhewei Decoction and western medicine groups began to ate more food,and feces of sparse and dry stools were less than before,and their mental and activity status was improved.2.Weight: There was no difference in the weight of each group of rats before modeling(P > 0.05).After modeling,the body weight of modeling groups was significantly lower than that of the normal control group,and the difference was significant(P < 0.01).After six weeks of drug intervention,compared with the model group,the weight gain of rats in each group was not significant,and the growth rate of each dose group of Jianpigubenhewei Decoction was slightly higher than that of the domperidone group,but there was no significant difference(P > 0.05).3.Blood glucose: The blood glucose of each group of rats were significantly higher than those in the normal control group at 3 days,1 week,6 weeks after the establishment of the model and 6 weeks of drug intervention(P < 0.01).After six weeks of drug intervention,compared with the model group,the blood glucose of rats in the high-dose group of Jianpigubenhewei Decoction decreased significantly(P < 0.01),while there was no significant difference in the other groups(P > 0.05).4.Gastric residual rate: The gastric residual rate in the high-dose group,middle-dose group,low-dose group of Jianpigubenhewei Decoction,domperidone group and model group were higher than that in the normal group(P < 0.05).The gastric residual rate of the high-dose group of Jianpigubenhewei Decoction was significantly lower than that of the model group(P < 0.05).5.Contents of serum MTL,GAS and Ghrelin: The contents of serum MTL and GAS in DGP model group was significantly higher than that in normal group,while the contents of serum Ghrelin decreased significantly(P < 0.01).The serum levels of MTL and GAS in each dose group of Jianpigubenhewei Decoction and domperidone group were lower than those in DGP model group(P < 0.01).The serum levels of Ghrelin in high,middle dose group of Jianpigubenhewei Decoction and domperidone group were significantly higher than those in DGP model group(P < 0.01).There was no significant difference in MTL and GAS content between high dose group of Jianpigubenhewei Decoction and domperidone group(P > 0.05).The serum Ghrelin levels in the high and middle dose groups of Jianpigubenhewei Decoction were slightly lower than that in domperidone group,but there was no significant difference(P > 0.05).6.Expression of ICC in gastric antrum: The content of ICC in gastric antrum in DGP model group was significantly lower than that of normal control group(P < 0.01).Compared with the model group,the ICC content in the antrum of rats in the high,middle dose of Jianpigubenhewei Decoction and domperidone groups increased significantly(P < 0.01).The ICC content in gastric antrum of rats in high and middle dose groups of Jianpigubenhewei Decoction was slightly lower than that in western medicine groups,but there was no significant difference(P > 0.05).7.Expression of SCF and c-kit protein in gastric antrum tissue: The expression of SCF and c-kit protein in gastric antrum of DGP model group was significantly lower than that in normal group(P < 0.01).Compared with model group,the expression of SCF protein and c-kit protein in gastric antrum tissue of rats in high,medium dose group of Jianpigubenhewei Decoction and domperidone group were significantly increased(P < 0.05).Conclusion: 1.Jianpigubenhewei Decoction can improve the general condition of diabetic gastroparesis rats,reduce gastric residual rate and serum MTL,GAS contents of DGP rats,increase the content of serum Ghrelin,promote gastric emptying in DGP rats and improve gastric motility in DGP rats.2.Jianpigubenhewei Decoction could up-regulate the expression of SCF/c-kit signaling pathway in gastric antral tissue of DGP rats,improve the functional activity of ICC,increase the content of ICC,improve gastric motility in DGP rats and promote gastric emptying in DGP rats.3.The effects of Jianpigubenhewei Decoction on promoting gastric emptying,improving gastric motility and regulating hormone may be related to SCF/c-kit signaling pathway.
