KdPT Ameliorates Imiquimod-induced Psoriasis-like Inflammation Of BALB/c Mice

Objective:To investigate the effect of Kd PT on the treatment of psoriatic mice by establishing a cell inflammatory model to study the anti-inflammatory activity of lepsul peptide acetate(Kd PT)in vitro and(IMQ)induced to establish a mouse psoriasis model.Methods and results:The effect of Kd PT on cell proliferation inhibition was detected by CCK8 method in vitro.GRIESS method was used to determine the content of NO in supernatant after LPS stimulation of mouse macrophages.ELISA detect the changes of inflammatory factors secreted by cells.The results showed that Kd PT had some inhibitory effect on the proliferation of skin-related cells such as Haca T 、Huvec S in the corresponding concentration.and present a certain drug dose dependence.The effect of the Kd PT on the related inflammatory factors showed that it had inhibitory effect on LPS induced Raw264.7 cells to secrete a large number of NO、IL-6 and other inflammatory factors and showed a certain drug dose dependence.The animal in vivo studies used imiquimot(IMQ)to establish a psoriasis symptom model.the clinical severity of the psoriasis was scored from the point of view of erythema,scales,and thickness of the mouse skin.HE staining,immunohistochemical analysis,q-PCR and other methods were taken to evaluate the efficacy of the in psoriasis.According to the results of previous experiments Kd PT gavage and intraperitoneal injection in the treatment of colitis has a certain therapeutic effect,but the previous period did not clearly study which way of treatment is better,so on the basis of the comparison of the treatment of psoriasis in mice.A study found that the gavage treatment group had better effect on IMQ induced psoriasis mice symptoms.Then by gavage to increase the dosage of drugs,the Kd PT administration group can alleviate IMQ induce psoriasis mice erythema,scales and other symptoms in a concentration-dependent reduction of IMQ spleen index.the HE staining also found that when increasing drug concentration,the thickness of mouse epidermis and the degree of keratinization insufficiency were reduced in different degrees compared with IMQ group.the expression of related inflammatory factors was detected by immunohistochemistry and q-PCR.the results also showed that Kd PT could significantly inhibit the expression of inflammatory related proteins.Conclusions:In this study,we found that Kd PT has a significant inhibitory effect on inflammatory factors in vitro and also has a good value-added inhibition on related skin cells.Through the IMQ induction psoriasis model,in the study through the treatment of drugs,respectively in the observation of skin pathological changes,spleen index determination,PASI score,skin tissue inflammation related protein changes,the results showed that Kd PT well alleviated psoriasis related symptoms.to sum up,Kd PT has a good anti-inflammatory activity in vitro and also relieves psoriasis symptoms in vivo,so this study hopes to provide a theoretical basis for the development of clinical drugs.
