Effect Of HMGB1 On Autophagy And Apoptosis Of Lymphocytes In Patients With Acute Pancreatitis

Abstract/Summary:
Objective: To investigate the expression of HMGB1 in peripheral blood of patients with acute pancreatitis(AP)and its effect on autophagy and apoptosis of lymphocytes.Methods: 76 patients with AP were admitted,including 28 patients with mild acute pancreatitis(MAP),25 patients with moderate severe acute pancreatitis(MSAP),23 patients with severe acute pancreatitis(SAP),and 25 healthy volunteers were admitted as control group.RT-q PCR was used to detect high mobility group protein B1(HMGB1),microtubule associated protein 1 light chain 3-II(LC3-II)and Caspase-3 in peripheral blood lymphocytes of AP patients.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum HMGB-1,LC3-II,and Caspase-3 protein levels in patients with AP.The autophagy rate and apoptosis rate of peripheral blood lymphocytes in AP patients were detected by flow cytometry.Conclusions:(1)The level of HMGB1 m RNA and serum protein in SAP patients was higher than that in control group,MAP group and MSAP group(P < 0.05);the level of HMGB1 m RNA and serum protein in MSAP group was higher than that in control group and MAP group(P < 0.05);the level of HMGB1 m RNA and serum protein in MAP group was higher than that in control group(P < 0.05).(2)The levels of Caspase-3,LC3-II m RNA and serum protein in SAP group were higher than those in control group,MAP group and MSAP group(P < 0.05);the levels of Caspase-3,LC3-II m RNA and serum protein in MSAP group were higher than those in control group and MAP group(P < 0.05);the levels of Caspase-3,LC3-II m RNA and serum egg white in MAP group were only slightly higher than those in control group(P > 0.05).(3)The autophagy rate and apoptosis rate of peripheral bloodlymphocytes in SAP group were higher than those in control group,MAP group and MSAP group(P < 0.05);the autophagy rate and apoptosis rate of peripheral blood lymphocytes in MSAP group were higher than those in control group and MAP group(P < 0.05);the autophagy rate and apoptosis rate of peripheral blood lymphocytes in MAP group were only slightly higher than those in control group(P > 0.05).(4)In SAP group and MSAP group,serum HMGB1 protein level was positively correlated with lymphocyte autophagy rate and apoptosis rate(P < 0.05).With the increase of HMGB1 level,lymphocyte autophagy rate and apoptosis rate increased.In SAP group and MSAP group,serum HMGB1 level was positively correlated with LC3-II and caspase-3 levels(P < 0.05).With the increase of HMGB1 level,LC3-II and caspase-3 levels increased.Results: 1.The expression level of HMGB1,autophagy and apoptosis of lymphocytes increased significantly in AP patients.2.High expression of HMGB1 can up regulate the expression level of autophagy related gene LC3-II and apoptosis related gene Caspase-3,which may be an important reason for the increase of autophagy and apoptosis intensity of peripheral blood lymphocytes in AP patients.
