Objective:To study the expression of DJ-1 in prostate cancer tissue and explore the relationship between DJ-1 and prostate cancer.To investigate the effect of DJ-1 on the proliferation and invasion of prostate cancer LNCa P cells.Method:(1)Tissue samples of 51 patients with prostate cancer were selected as the experimental group and 50 patients with benign prostatic hyperplasia as the control group.All the patients in the experimental group were newly diagnosed and with no endocrine therapy or radiotherapy and chemothertherpy treatment,and were divided into subgroups according to Gleason score(Gleason score ? 6,Gleason score = 7,Gleason score ? 8).Immunohistochemistry SP method was used to detect the expression of DJ-1 in 50 cases of BPH and 51 cases of prostate cancer.The correlation between the expression level of DJ-1and the clinical and pathological parameters of prostate cancer was analyzed.The difference between the expression level of DJ-1 and the methylation modification level and prostate cancer and adjacent tissues was explored by TCGA database.(2)LNCa P cell lines were cultured and transfected into three groups: CON,NC and KD.The transfection efficiency was tested by green fluorescence,and the silencing efficiency was verified by Western blot and RT-q PCR.Then the proliferation and invasiveness of prostate cancer cells were detected by CCK-8 technology,plate cloning technology,Transwell chamber experiment and cell scratch experiment.Results :(1)The expression level of DJ-1 in prostate cancer(IRS: 3.7 ± 0.51)was significantly higher than that in prostatic hyperplasia(IRS: 2.2 ± 0.57),the difference was statistically significant(P < 0.05);Gleason score ? 8 group IRS: 7.9 ± 0.9,Gleason score = 7group IRS: 4.7 ± 0.6,Gleason score ? 6 group IRS: 2.7 ± 0.6,the difference was statisticallysignificant(P < 0.05),There was a positive correlation between the expression level of DJ-1and the Gleason score of prostate cancer.There was significant correlation between the expression of DJ-1 and prostate cancer stage,Gleason score,TPSA value and the number of metastasis(P < 0.05).The expression of DJ-1 m RNA in prostate cancer tissues was significantly higher than that in normal tissues,while the level of methylation was significantly lower than that in normal tissues(P < 0.05).Survival analysis showed that the expression of DJ-1 was significantly correlated with the biochemical recurrence of PCA(P =0.032).(2)After 72 hours of lentivirus infection,the green fluorescence test showed that the transfection efficiency was over 80%.Western blot and RT-q PCR analysis showed that the expression level of DJ-1 m RNA and protein in KD group was significantly lower than that in CON group and NC group(P < 0.01),but there was no significant difference between CON group and NC group(P > 0.05).CCK-8 experiment showed that the cell growth rate of KD group was significantly lower than that of NC group and CON group(P < 0.01)over time;the cell proliferation and clone formation rate of KD group was significantly lower than that of NC group and con group(P < 0.05);Transwell showed that the number of invasive cells of KD group was significantly lower than that of CON group and NC group(P < 0.01),and there was no statistical difference between CON group and NC group The cell scratch test showed that the scratch healing degree of KD group(12.26%)was significantly lower than that of con group(49.28%)and NC group(47.88%)(P < 0.001).Conclusions:DJ-1 is significantly up-regulated in prostate cancer,positively correlated with Gleason score,pathological stage,TPSA value and number of metastasis,and the high expression of DJ-1 is related to biochemical recurrence of prostate cancer;silence of DJ-1 can significantly reduce the proliferation,invasion and migration of LNCa P cells.DJ-1 may be a new tumor marker and a new rake point for prostate cancer treatment.
