The Expression And Clinical Significance Of MiR-152 And MiR-182 In Endometrial Tissue

Objective: To detect the differential expressions of miR-152 and miR-182 in normal endometrial tissues,endometrial atypical hyperplasia tissues and endometrial cancer tissues,analyze the relationships between the two genes and clinicopathological parameters of endometrial cancer,and assessment the valueofdiagnosisfor endometrial cancerpreliminarily.Method: 110 patients who underwent surgical treatment in Affiliated Hospital of Hebei University from September 2017 to December 2019 were selected,including 60 patients with endometrial cancer(47 cases of endometrial adenocarcinoma,13 cases of special endometrial cancer),20 patients with endometrial atypical hyperplasia and 30 patients with uterine fibroids or uterine prolapse.Fresh endometrial lesions and normal tissues were cut out during the operation.Real-time fluorescence quantitative PCR(RT-q PCR)was used to detect the expression of miR-152 and miR-182 in normal endometrial tissues,endometrial dysplasia tissues and endometrial cancer tissues,respectively.The differences of the relative expressions of the two genes in normal tissues and cancer tissues were analyzed separately.And the relationships between the relative expressions of the two genes and the clinicopathological characteristics of endometrial cancer were analyzed separately,too.The changes of the two genes from the group of normal endometrium to the group of endometrial dysplasia to the group of endometrial adenocarcinoma wereanalyzedfurtherly.Drawing receiver operating characteristic curve(ROC),calculating sensitivity,specificity and area under the ROC curve(AUC)to evaluate their diagnostic efficacy for endometrial cancer.Result: The result of real-time fluorescence quantitative PCR showed:1.The relative expressions of miR-152 in normal and cancer specimens of the endometrium were 0.999±0.416 and 0.583±0.226,respectively.The relative expression in cancer tissues was significantly lower than that in normal tissues,and the difference was statistically significant(P<0.05).miR-152 showed lower expression levels in endometrial cancer patients with stage ?-?,myometrial invasion depth ? 1/2,vascular space invasion,and positive lymph node metastasis.And its expression in adenocarcinoma patients was alsolower than that of special endometrial cancer patients,the differences were statistically significant(P<0.05).However,the relative expression differences in patients with different pathological grades,ages and menopausal states were not statistically significant(P>0.05).2.The relative expressions of miR-152 in patients with normal endometrial,endometrial dysplasia,and endometrial adenocarcinoma were 0.999±0.416,0.714±0.239 and 0.627±0.214,respectively,showing a gradual downward trend.And the relative expression of the normal group was lower than that of the endometrial dysplasia group and the endometrial adenocarcinoma group,and the differences were statistically significant(P<0.05).However,there was no statistically significant difference in the relative expressions between the endometrial dysplasia group and the endometrial adenocarcinoma group(P>0.05).3.The relative expressions of miR-182 in normal and cancer specimens of the endometrium were 1.356±0.709 and 3.301±1.445,respectively.The relative expression in cancer tissues was significantly higher than that in normal tissues,and the difference was statistically significant(P<0.05).The expression level of miR-182 was higher in patients with special types of endometrial cancer than that in patients with adenocarcinoma,and it was higher in patients with stage III-IV of endometrial cancer than that in patients with stage I-II,and the difference was statistically significant(P<0.05).The relative expressions of this gene were not statistically significant in patients of different ages,pathological grades,and muscle depths.And they were also not statistically significant in patients with or without vascular space invasion,lymph node metastasis,and menopause(P>0.05).4.The relative expressions of miR-182 in normal endometrium,endometrial dysplasia,and endometrial adenocarcinoma were 1.356±0.709,1.827±0.422 and 3.036±1.309,respectively,showing a gradual increase trend,and the difference was statistically significant(P<0.05).And comparing the expressions of miR-152 between the groups pairwise showed a statistical significance(P<0.05).The ROC curve result showed:1.Taking the normal endometrial group as a reference,miR-152 had an area under the ROC curve of 0.810,a sensitivity of 86.7%,and a specificity of 73.3% in the diagnosis ofendometrial cancer.2.Taking the normal endometrial group as a reference,miR-182 had an area under the ROC curve of 0.932,a sensitivity of 93.3%,and a specificity of 86.7% in the diagnosis of endometrial cancer.Conclusion:1.miR-152 and miR-182 were abnormally expressed in endometrial cancer tissues,which might be tumor suppressor genes and tumor-promoting genes of endometrial cancer,respectively.The relationships between the two genes and clinicopathological characteristics suggested that they had a certain value in diagnosing and evaluating the malignancy and clinical progress of endometrial cancer.2.Both miR-152 and miR-182 had good diagnostic efficiency for endometrial cancer,and they were expected to become molecular biological markers for early diagnosis of endometrial cancer.
