Abstract/Summary:
Objectives:Non-Small Cell Lung Cancer(NSCLC)databases from The Cancer Genome Atlas(TCGA)and tissue chips were used to explore the expression of AXL,Vimentin and E-cadherin and their relationship with clinicopathologic parameters and prognosis,to explore the mechanism of AXL in the occurrence and development of lung cancer,and to provide a new idea for targeted therapy of NSCLC.Method:1.To screen 500 cases of Lung Squamous Cell Carcinoma and 512 cases of Lung Adenocarcinoma with complete data by downloading TCGA Lung Squamous Cell Carcinoma and Lung Adenocarcinoma database,to analyze the gene expression of AXL,Vim,CDH1 and their relationship with clinicopathologic parameters and prognosis.2.The expression levels of AXL in TCGA Lung Adenocarcinoma database was divided into high and low expression groups,and Gene Set Enrichment Analysis(GSEA)was used to study the gene set of biological pathway affected by AXL expression level.3.The expression of AXL,Vimentin and E-cadherin in NSCLC tissue microarray and the expression of AXL in lung disease spectrum tissue microarray were analyzed by immunohistochemistry.Results:1.Analysis of the TCGA Lung Squamous Cell Carcinoma and Adenocarcinoma of the Lung database revealed:AXL in Lung Squamous Cell Carcinoma was lower than that in normal lung tissue and higher than that in Lung Adenocarcinoma.By Spearman’s method,we found that AXL and VIM were positively correlated with each other in Lung Squamous Cell Carcinoma and Adenocarcinoma(P<0.05);both VIM and CDH1 were negatively correlated in Lung Squamous Cell Carcinoma and Adenocarcinoma(P<0.05).AXL and CDH1 were negatively correlated in Lung Squamous Cell Carcinoma(P<0.05)and positively correlated in Lung Adenocarcinoma(P<0.05).The relationship between the expressions of AXL,VIM and CDH1 and clinicopathologic parameters was analyzed.The results showed that high expression of AXL associated with VIM,high expression of AXL associated with CDH1,low expression of AXL associated with CDH1,high expression of VIM and high expression of VIM associated with low expression of CDH1 promoted the clinical staging and progression of tumors.A univariate and multivariate analysis of NSCLC outcomes showed:T staging,N Staging,M staging,TNM staging,AXL expression,AXL overexpression+VIM overexpression,AXL overexpression + CDH1 underexpression,AXL overexpression +VIM overexpression+ CDH1 underexpression decreased overall survival of NSCLC patients.Moreover,M staging,AXL expression,AXL overexpression+Vim overexpression,AXL overexpression + CDH1 low expression,AXL overexpression +VIM overexpression + CDH1 low expression were independent prognostic factors of NSCLC.In addition,AXL overexpression affected the overall survival of patients with lung Adenocarcinoma,but had no significant effect on the prognosis of patients with Lung Squamous Cell Carcinoma.2.According to GSEA enrichment analysis,AXL overexpression may be involved in the progression of lung cancer through biological processes such as Inflammation and Extracellular Matrix and related pathways.3.Tissue microarray analysis of NSCLC revealed:the expressions of AXL,Vimentin and E-cadherin in NSCLC were higher than those in normal lung tissues.AXL and Vimentin showed a positive correlation by analyzing the correlation of protein expression.When AXL and Vimentin expression were analyzed in relation to clinicopathologic parameters,AXL was found to be overexpressed in tissues with lymph node metastasis compared with those without.4.The expression of AXL protein in lung benign diseases was higher than that in normal lung tissues.Conclusion:1.According to the expression of AXL,Vimentin and E-cadherin in NSCLC and their relationship with clinicopathologic parameters,AXL may interact with E-cadherin to maintain EMT process,thus promote the invasion and poor prognosis of lung cancer.2.The GSEA enrichment analysis found that AXL promotes the development of lung cancer through biological processes and related pathways,such as Extracellular Matrix and Inflammation.3.Through the analysis of Lung Benign Disease tissue microarray,it was found that AXL protein was overexpressed in inflammatory disease.
