Objective: Taking Chinese healthy adult subjects as the research object,the effects of bear bile powder and artificial bear bile powder on healthy human intestinal flora were studied,and the subjects were identified with traditional Chinese medicine constitutions to explore the intestinal bacteria between different constitutions of traditional Chinese medicine The differences in groups provide theoretical support for the rational application of bear bile powder,and provide a reference for further research on whether different constitutions of traditional Chinese medicine have specific flora.Methods: A randomized controlled trial was used.From July 2019 to November 2019,a total of 48 subjects who met the inclusion and exclusion criteria were charged in the Phase I clinical ward of the Second Affiliated Hospital of Tianjin University of Traditional Chinese Medicine,randomly divided into experimental group A,experimental group B,and control group,each Group 16 cases,recorded general information,physical examination,blood routine,urine routine,stool routine,biochemical,coagulation,electrocardiogram,chest radiography,ultrasound(hepatobiliary pancreas spleen).The experimental group A was given bear bile powder intervention,the experimental group B was given artificial bear bile powder intervention,and the control group was given placebo for 14 consecutive days.It was observed that 72 hours after the last medication,the subjects were provided with a prescribed diet in the ward.Tea,coffee and beverages are not allowed during the test.Participants retained a portion of feces before the intervention and after the last medication.The microbial16 S amplicon absolute quantitative test was used to detect fecal flora,and the differences in composition of intestinal flora before and after medication were analyzed and compared.Results:At the phylum level,the relative abundances of Bacteroidetes,Actinobacteria,Verrucomicrobia,Verrucomicrobia(P<0.05),and Proteobacteria(Proteobacteria)in the bear bile powder group decreased.Abundance increased(P<0.05),the relative abundance of Bacteroides phylum in the artificial bear bile powder group was significantly reduced(P<0.05),and the relative abundance of Pachycephalum and Fusobacterium was significantly increased,with statistical difference(P<0.05)),there is no significant difference in the inhibitory effect of bear bile powder and artificial bear bile powder on Bacteroidetes(P>0.05).At the class level,the relative abundance of Bacteroidia(Bacteroides),Actinobacteria(Bacteroides),Betaproteobacteria(Beta-Proteobacteria),and Verrucomicrobiae(Verrucomicrobia)were significantly reduced(P<0.05),The relative abundance of Gammaproteobacteria(γ-proteobacteria)increased(P<0.05);the relative abundance of Bacteroides and β-proteobacteria in the artificial bear bile group decreased significantly(P<0.05),Clostridia and Clostridium The relative abundances of the class and erysipelas hyphae increased significantly(P<0.05).At the mesh level,the relative abundances of Bacteroidales,Bacteroidales,Burkholderiales and Burkholderiales were significantly reduced(P<0.05),Enterobacteriales(Enterobacteriales)),Fusobacteriales(Clostridia),Bacillales(Bacillus),the relative abundance increased significantly(P<0.05);the artificial bear bile powder group Bifidobacteriales(Bifidobacteria),the relative abundance was significantly reduced,Enterobacteria,Clostridium The relative abundance of the order,Bacillus order increased significantly(P<0.05).At the family level,the relative abundance of the Ruminococcaceae(Bacteroides family)and Bifidobacteriaceae(Bifidobacteriaceae)groups in the bear bile powder group were significantly reduced,and Lachnospiraceae(Enterobacteriaceae),Fusobacteriaceae(Fosobacteriaceae))The relative abundance was significantly increased(P<0.05);the relative abundance of Bifidobacteriaceae in the artificial bear bile group was significantly reduced(P<0.05),and the relative abundance of Streptococcaceae was significantly increased(P<0.05).At the genus level,the relative abundance of Bifidobacterium in the bear bile powder group was significantly reduced(P<0.05),and the relative abundance of Escherichia was significantly increased(P<0.05);the artificial bear bile powder group The relative abundance of Bifidobacterium decreased(P<0.05),and the relative abundance of Escherichia increased(P<0.05).At the species level,the bear gall powder group Bacteroides uniformis(Bacteroides EA1509E)Increase(P<0.05);the relative abundance of Streptococcus_salivarius(Streptococcus salivarius)and Lachnospira_pectinoschiza(P.pectinosarum)in the artificial bear bile powder group(P<0.05),Escherichia coli_K-12(E.coli K-12 line)Abundance increased(P<0.05).In this study,46 healthy subjects were identified by TCM constitution identification,37 cases(80.43%)were calm quality,4 cases(8.70%)were yin deficiency,2 cases(4.35%)were qi depression,and 1 case was yang deficiency.(2.17%),one case with qi deficiency(2.17%)and one case with blood stasis(2.17%).Compared with the Yin deficiency group,at the portal level,the relative abundance of the Parcubacteria group was lower than that of the Yin deficiency group(P<0.05);at the department level,the Listeriaceae(The relative abundance of Listeria monocytogenes group is lower than that of Yin deficiency group(P<0.05);at the genus level,the relative abundance of Coprococcus(Pectococcus)in the peace group is higher than that of Yin deficiency group(P<0.05);At the species level,the relative abundance of Parabacteroides distasonis(P.bacterium)is higher than that of Yin deficiency group(P<0.05).Conclusion:1.Bear bile powder and artificial bear bile powder have no significant effect on intestinal flora alpha diversity.2.Bear bile powder against Bacteroidetes,Bacteroides,Actinomycetes,β-proteobacteria,Verrucobacteria,Bacteroides,Burkholderia,Verrucobacteria,Bifidobacterium Bacilliaceae,Bifidobacterium,Bacteroides monoforme,Eubacterium rectum ATCC 33656,Bacteroides feces,Streptococcus salivarius,Bacteroides ellipsoidis,Bifidobacterium pseudochain etc.have an inhibitory effect.3.Artificial bear bile powder has an inhibitory effect on Bacteroides,Bacteroides,β-proteobacteria,Bifidobacteria,Bifidobacteriaceae,Bifidobacterium,Streptococcus salivarius,Trichosporon spp.,etc..4.Bear bile powder and artificial bear bile powder have a wide range of inhibitory effects on probiotics,neutral bacteria and pathogenic bacteria.The specific inhibitory types of the two are slightly different,and may indirectly promote the growth of certain bacterial groups.5.The relative abundances of the population of Pingjian group,Listeria monocytogenes,Faecoccus spp.,and Gibberella spp.in the peace-quality population and the yin-deficiency population are different.
