Objective:To study the distribution characteristics of the four single nucleotide gene polymorphisms(SNPs)of Nur77 genes rs10876228,rs11169989,rs1283155,and rs2242107 in healthy people in Guangxi,and investigate its distribution characteristics in diflent gender and races.To study the relationship between Nur77 gene rs10876228,rs11169989,rs1283155,rs2242107 polymorphisms and susceptibility and severity of sepsis.The serum levels of Nur77 in sepsis group and the control group were detected.Methods:A total of 210 healthy people from Guangxi were recruited in this study.SNPs of Nur77 genes rs10876228,rs11169989,rs1283155,and rs2242107 were detected using SNa Pshot minisequencing.The Nur77 gene polymorphism of the Guangxi population were compared with that of the American,South Asian,European,African,and East Asian populations which were released by the 1000 genomes project.A total of 207 patients with sepsis(sepsis group)and 210 healthy subjects(control group)were enrolled in the study.Genotype of rs10876228,rs11169989,rs1283155,rs2242107 SNPs were determined by a SNa Pshot Multiplex sequencing assay.The sepsis group were subsequewtly divided into sepsis non-shock group and septic group.And Explore the difference between its distribution and healthy people.Nur77 gene polimorphidms and serum Nur77 levels were compared between healthy group and sepsis group,and subsequently,sepsis without shock group and septic shock group.Serum Nur77 levels in all subjects were determined by ELISA.To investigate the relationship between Nur77 polymorphisms and severity of sepsis.Results:(1)There were no statistically significant differences in the distribution of allele and genotype of the Nur77 genes rs10876228,rs11169989,rs1283155,and rs2242107 between different genders in the Healthy peoples in Guangxi population(all P > 0.05).(2)There are three genotypes of CC,CT and TT in rs10876228 in Guangxi population,the frequencies are 18.6%,52.4% and 29.0% respectively,and the frequencies of C and t allele are 44.8% and 55.2% respectively.The frequency of rs10876228 genotype and allele in Guangxi population were statistically significant(all P < 0.01)compared with that of American,South Asian,European and African populations,but there was no statistical significance(P > 0.05)when compared with East Asian population.(3)There are three genotypes of AA,AC and CC in rs11169989 in Guangxi population,the frequencies are83.3%,16.2% and 0.5% respectively,and the frequencies of a and C allele are 91.4% and8.6% respectively.The frequency of rs11169989 genotype and allele were statistically significant(all P < 0.05)compared with American population,but there was no statistically significant(P > 0.05)compared with South Asian and European population.There was no significant difference in the genotype of rs11169989 between Healthy peoples in Guangxi population and African and East Asian population(P > 0.05),but there was significant difference in rs11169989 allele between Guangxi population and African and East Asian population(all P < 0.01).(4)There are three genotypes of AA,GA and GG in rs1283155 in Guangxi population,the frequencies are 13.8%,40.5% and 45.7% respectively,and the frequencies of A and G allele are 34.0% and 66.0% respectively.There were significant differences in the frequency of rs1283155 genotype and allele between Guangxi population and American,South Asian,European,African and East Asian(all P < 0.01).(5)There are three genotypes of AA,GA and GG in rs2242107 in Guangxi population,the frequencies are18.1%,46.2% and 35.7% respectively,and the frequencies of A and G allele are 41.2% and58.8% respectively.There were significant differences in rs2242107 genotype and allele frequency between Guangxi population and South Asian,European,African and East Asian population(all P < 0.01),but there was no significant difference between Guangxi population and American population(P > 0.05).(6)There were three genotypes of CC,CT and TT inrs10876228 in the control group and sepsis group,and there was no significant difference in the frequency of each genotype under various genetic models between the two groups(all P >0.05).(7)There were three genotypes of rs11169989,AA,AC and CC in the control group,and there were AA and AC genotypes in sepsis group.There was no significant difference in the frequency of each genotype under different genetic models between the two groups(all P > 0.05).(8)There were three genotypes of AA,GA and GG in rs1283155 in the control group and sepsis group,and there was no significant difference in the frequency of each genotype under various genetic models between the two groups(all P > 0.05).(9)There were three genotypes of AA,GA and GG in rs2242107 in the control group and sepsis group,and there was no significant difference in the frequency of each genotype under various genetic models between the two groups(all P > 0.05).(10)There was no significant difference in the genotype and allele frequency of Nur77 polymorphism between sepsis group and control group(all P > 0.05).(11)The genotype and allele of the rs10876228?rs11169989?rs1283155and rs2242107 of Nur77 genes in sepsis non-shock group were not significantly different between different genders(all P > 0.05).(12)The genotype and allele of the rs10876228rs11169989?rs1283155 and rs2242107 of Nur77 genes were not statistically different in the septic shock group between different genders(all P > 0.05).(13)The genotypes and alleles of the rs10876228 rs11169989?rs1283155 and rs2242107 of Nur77 genes was no statistically significantly different between sepsis without shock group and septic shock group(all P >0.05).(14)Serum Nur77 level in patients with sepsis was lower than that of control group(P<0.05).there was no statistically significantly differele in serum Nur77 levels between the sepsis non-shock group and septic shock group(P > 0.05).Conclusion:(1)The Nur77 genes rs10876228,rs11169989,rs1283155,and rs2242107 are polymorphic in healthy people in Guangxi,and there is no significant in genders difference.(2)There are significant genetic differences between Guangxi population and American,South Asian,East Asian,European and African population in the Nur77 genesrs10876228,rs11169989,rs1283155 and rs2242107,which may lead to a significant difference in the incidence and clinical manifestations of certain diseases among them.(3)The Nur77 genes rs10876228,rs11169989,rs1283155,and rs2242107 were genetically polymorphic in the sepsis group,but failed to play a role in susceptibility to sepsis.(4)The Nur77 may be used as a diagnostic marker for sepsis,but it cannot predict the severity of sepsis.
