Research purposes: old people with old legs,aging of leg muscles will lead to unstable support,inconvenient walking,it is easy to fall.Do not pay attention to the exercise of the muscles of the elderly,they will be older,and the standard of living will be greatly reduced Long-term aerobic exercise,the balance ability,coordination ability and agility ability of the elderly are greatly improved,and the daily living ability of the elderly is greatly improved.This study explores the operation of the aged rats through exercise intervention.The expression of igf-1 and mstn protein in skeletal muscle of aged rats in order to provide reference and reference for domestic basic research and clinical application research in this field.Research methods: In this experiment,40 male SD rats of 4 weeks old were selected and routinely raised to 8 weeks old and weighed 350g~400g.,were established by subcutaneous injection of D-galactose 0.05mL/50 g to establish a rat model of aging.The rats were divided into control group(C),exercise group(E)aging group(D)exercise inhibition aging group(A)aging exercise group(DE),five groups,8 groups each Animals caged,free feeding and drinking,Sports model All of them used moderate-intensity aerobic exercise.After the grouping,the control group did not exercise.All the exercise models were trained for one week,and the speed of the running platform was gradually accelerated to gradually adapt to the training intensity of the experimental design.The running platform slope was set to 0.The speed of the treadmill finally reaches 1.1m/min,the duration is from 20 min to 60 min,and the training is 6 days a week for 8 weeks.After the ninth week of the water maze test,the rat gastrocnemius muscle was quickly removed and placed in pre-cooled physiological saline to remove connective tissue such as fat.The filter paper was blotted,wrapped in tin foil,stored in liquid nitrogen,and finished.After the samples were all stored in a-80 ° C refrigerator,after tissue grinding,the concentration was measured by BCA kit,and the expression of IGF-1 and MSTN protein was detected by Western blotting.result:Western blot protein detection1.The expression of IGF-1 protein from strong to weak was E group>C group>A group>DE group>D group,the exercise group(E)rat skeletal muscle IGF-1 protein expression was the most,and the control group(C)was significantly different(P<0.05);aging model group(D)had the least expression of IGF-1 protein in skeletal muscle of rats,and it had a very significant difference compared with the control group(C)(P)<0.01);In addition,the expression of IGF-1 protein in skeletal muscle of aging exercise group(DE)was significantly different from that of exercise-suppressed aging group(A)(P<0.01);aging exercise group(DE)was large.The expression of IGF-1 protein in skeletal muscle of rats was significantly different from that in aging model group(D)(P<0.05).2.The expression of MSTN protein from strong to weak was D group>DE group>A group>C group>E group.The aging model group(D)had the most expression of MSTN protein in skeletal muscle of rats,and it was compared with the control group(C).There was a very significant difference(P<0.01);the expression of MSTN protein in the skeletal muscle of the exercise group(E)was the least,and there was a significant difference compared with the control group(C)(P<0.05);The expression of MSTN protein in skeletal muscle of aging exercise group(DE)was significantly different from that of exercise-suppressed aging group(A)(P<0.01).The expression of MSTN protein in skeletal muscle of aging exercise group(DE)rats The aging model group(D)had a very significant difference(P<0.01).in conclusion:1.The expression of IGF protein in rat skeletal muscle decreased with age.The treadmill exercise can effectively increase the expression of IGF-1 protein in rats,and it is suitable for aged rats.In addition,during accelerated aging,exercise can significantly slow down the bones.Decreased muscle IGF-1.2.The expression of MSTN protein in rat skeletal muscle increased with age.The expression of MSTN protein in rats was significantly decreased by exercise intervention for 8 weeks.The expression of MSTN protein was decreased in old rats through exercise,and during the process of accelerating aging,exercise It can significantly inhibit the rise of MSTN protein.
