Objectives:To offer a theoretical basis for the metabolic syndrome diseases,this study used normal and ovariectomized obese rats to observe the effects of treadmill exercise on body weight,vital visceral weight,weight index,serum Irisin level,and the effect of FNDC5 and its related molecular protein expression.Methods: Forty-five healthy non-pregnant female SD rats of three-month-old were randomly divided into sham-operation group(Sham n = 20)and ovariectomized group(OVX n = 25)according to their body weight.After the sham and OVX surgical operation,sham group was divided into sham resting group and sham-E group,as well as,OVX group was divided into OVX resting group and OVE-E group.Three weeks after surgery,both the sham-E and OVE-E rats were trained for a period of 8 weeks(45min/day,5 uphill,18m/min,5 times/week).Rats were anesthetized with 10% chloral hydrate(3ml/kg)within 24-36 hours after the last exercise intervention.After the end of the study,the blood lipids were measured by 721,the level of Irisin was detected by ELISA,the AMPK?,p-AMPK?,PGC-1? and FNDC5 protein expression of skeletal muscle and myocardium were detected by Western blot.The results of the experiment:(1)The body weight in sham-E group were significantly lower than sham resting group at the 6th and 7th weeks whill no difference at other times.The body weight in OVE-E group was significantly lower than OVE resting group during the last two weekswhill no difference at other times.(2)Compared with sham resting group,the serum Irisin level was no significant change in sham-E group.Compared with OVX resting group,the serum Irisin level was no significant change in ovariectomized exercise group.(3)In Skeletal muscle.Compared with sham resting group,the AMPK? and p-AMPK? protein expression of Long extensor muscle in sham-E group were decreased significantly,the PGC-1? protein expression was increased significantly,while no difference in the FNDC5 protein expression;Compared with OVX resting group,no difference in the AMPK? and p-AMPK? protein expression of Long extensor muscle in OVX-E group,the PGC-1? and FNDC5 protein expression were significantly increased.Compared with sham resting group,the AMPK? and PGC-1? protein expression of soleus musclein in sham-E group were significantly increased and the p-AMPK? and FNDC5 protein expression were significantly decreased;Compared with OVX resting group,there was no difference in the AMPK? and PGC-1? protein expression in OVX-E group,and the p-AMPK? and FNDC5 protein expression were significantly increased.(4)In myocardium,Compared the sham resting group,the AMPK?,p-AMPK?,PGC-1? and FNDC5 protein expression of myocardium in sham-E group were significantly decreased;Compared with OVX resting group,the AMPK?,PGC-1? and FNDC5 protein expression in OVX-E group were significantly increased,while no difference in the p-AMPK? protein expression.Experimental conclusion:(1)exercise can reduce body weight of OVX rats.(2)Exercise has no effect on serum Irisin levels in OVX rats.(3)exercise can Increase the PGC-1? and FNDC5 protein expression of long extensorium in OVX rats;can Increase the p-AMPK? and FNDC5 protein expression of soleus muscle in OVX rats.(4)Exercise can Increased the AMPK??PGC-1? and FNDC5 protein expression of Myocardium in OVX rats?...
