| Objective: The experimental subjects selected in this study wer e 6-hydroxydopamine hydrobromide(6-OHDA)Parkinson's rats,with re gular aerobic exercise as the intervention method.Regular aerobic ex ercise on 6-OHDA induced Parkinson's(PD)in the rat striatum syna ptic protein involved in synaptic plasticity(SYN),postsynaptic densit y protein-95(PSD-95)and calmodulin kinase(CaMK)signaling path way explore the synaptic plasticity related proteins induced by regul ar aerobic exercise for Parkinson's disease(PD)nerve protective effe ct.It provides experimental theoretical basis for the clinical treatment of PD.Methods: forty-eight SPF grade healthy male SD rats of 3 mon ths old were randomly divided into 4 groups,namely the quiet salin e group(SED)and the exercise saline group(EX).In the SED-PD group and the exercise PD group(EX-PD),the first 3 days of adapt ive training were conducted(the speed was 10m/min,10 min on the first day,20 min on the second day,25 min on the third day,and the sl ope was 0°).After a day off,follow an exercise regimen for 8 weeks.After the exercise intervention,the collective model building.After t he completion of modeling,all the rats recovered for 3-4 days,and the exercise group received 2 weeks of recovery training.Train five times a week,with wednesdays and Sundays off.Behavioral detection was conducted on the 7th and 14 th day after the completion of th e modeling,and striatum detection was conducted on four groups of rats 24 hours after the last behavioral detection.HE staining to obs erve changes of shape and structure of the brain striatum,immunohistochemical detection striatum alpha-synuclein(alpha-synuclein),Tyrosi ne hydrogenase(TH),the SYN,PSD-95,CaMKⅡalpha,the expression o f glutamate receptor(AMPAR)protein,RT-qPCR detection the striatu m the SYN,PSD-95,CaMKⅡalpha,AMPAR mRNA expression.Research results:(1)results of neurobehavioral rotation experiment:the disease in cidence of PD was 77.78% in the sed-pd group and 71.43% in the ex-pd group,respectively.(2)HE staining: The 6-OHDA injection sites of the experiment al group were all located on the injured side.The striatum of the S ED and EX groups of the rats could clearly see the dense band,the number of neuronal cells was large,the shape was intact,and the arr angement was well Orderly.In the SED-PD and EX-PD groups,the striatum is difficult to distinguish the dense band.The nerve damag e is obvious.The neurons and nerve fibers are degenerated,necrotic,and missing.(3)the IHC results show that compared with the corresponding resting between groups of rats,the rat striatum TH movement grou p significantly increased protein expression showed a trend of incre ase,the SYN,PSD-95 and CaMKⅡalpha,AMPAR protein expression i ncreased significantly,is on the rise,the EX and EX-PD group of al pha synuclein protein expression significantly reduced,a downward tr end.(4)the RT-qPCR results show that the movement to promote r at striatum SYN,PSD-95 and CaMKⅡalpha,AMPAR mRNA expressi on level increased;Compared with the SED group,SED-PD group in the rat striatum SYN,PSD-95,CaMKⅡalpha,AMPAR mRNA expressi on was significantly decreased(P<0.01);Compared with the EX group,the EX-PD group in the rat striatum SYN and CaMKⅡalpha,AMP AR mRNA expression was significantly decreased(P<0.01),the PSD-95 mRNA expression was significantly decreased(P<0.05);Compared with the SED-PD group,the EX-PD group in the rat striatum SYN,PSD-95,CaMKⅡalpha,AMPAR mRNA expression quantity increased significantly(P<0.05);Compared with SED group,the EX group in the rat striatum SYN mRNA expression quantity increased significantly(P<0.01),the PSD-95,CaMKⅡ alpha mRNA expression quantity incre ases slightly(P>0.05),AMPAR mRNA expression quantity increased s ignificantly(P<0.05).Conclusion: regular aerobic exercise intervention reduces the P D disease incidence rate,effectively relieve 6-OHDA toxic effect,to a certain extent,prevent and reduce the behavior of dysfunction in rat s,increase the related protein expression of SYN,PSD-95,and regula te CaMKⅡ alpha/AMPAR signaling pathways,improve the morpholog y and the function of the synaptic plasticity,and reduce the expressi on of brain striatum alpha synuclein,play to the regular aerobic exer cise of PD rat striatum nerve protective effect. |
