Objective: Research purposes: this study through movement joint ALDH2 agonist Alda-1 as a means of intervention,selected from 6-OHDA in PD model of SD rats as intervention object,explore aerobics joint ALDH2 agonist striatum of PD rats protein involved in synaptic plasticity CaMK??,Syn1,TH and alpha Syn and ALDH2,as well as the influence of PI3K/AKT/mTOR signaling pathways,such as,for the pathogenesis of PD and exercise rehabilitation,etc.Provide certain theoretical support for the clinical treatment.Object and methods: 92 healthy 3-month-old SPF male SD rats were selected,and after 2 weeks of adaptive feeding,they were randomly divided into 6 groups,each of which was a quiet control group(SED,n = 10);exercise Saline control group(EX,n = 10);Parkinson sedentary group(PD-SED,n = 18);Parkinson exercise runner group(PD-EX,n = 18);ALDH2-agonist Parkinson sedentary group(AG-PD-SED,n = 18);ALDH2-agonist Parkinson exercise runner group(AG-PD-EX,n = 18).Three days of adaptive training followed by 8 weeks of incremental load aerobic exercise.During the rearing period,rats(4 per cage)have free access to food and water.The indoor temperature is maintained at 22 ° C to 25 ° C,and the relative humidity is maintained at 45% to 55%.An alternating cycle of 12 hours light and 12 hours dark was used.After the training program,rats were modeled for one week with Parkinson's disease.Rats were injected with 6-OHDA into the striatum on the right side of the rat's brain.After two weeks of modeling,behavioral evaluation of the rotation experiment was performed.After 24 h of the last rotation experiment,the brain striatum tissue and serum samples were taken.The serum biochemical indicators of oxidative stress in rats were detected,and the expression of striatum ?-Syn,Syn1,TH,CaMK??,ALDH2,PI3 k / AKT / mTOR signaling pathways were detected by immunohistochemistry or real-time quantitative PCR technology.Results: the success rate of modeling was 77.8% in the pd-sed group,61.1% in the pd-ex group,72.2% in the ag-pd-sed group,and 66.7% in the ag-pd-ex group.Exercise combined with ALDH2 agonist significantly increased the activity of GSH in serum(P<0.05).Exercise combined with ALDH2 agonist significantly increased the expression of ALDH2 mRNA,Syn1 mRNA,and PI3K/AKT/mTOR mRNA(P<0.05).Compared with the non-agonist group,the mRNA expression of PI3K/AKT/mTOR in PD group treated with agonist was significantly different(P<0.01).IHC results show that the movement joint ALDH2 agonist intervention,reduce the PD rat striatum alpha the expression of Syn,improved the ALDH2,Syn1,CaMK??,TH protein expression,promote the activation of PI3K/AKT/mTOR signaling pathway.Research conclusions: Aerobic exercise combined ALDH2 agonist Alda-1 as a intervention factor,in eight weeks intervention experiment,significantly raised Syn1,CaMK??,TH and ALDH2 protein expression in rat striatum,and raised the PI3K/AKT/mTOR signaling pathway activity.Aerobic exercise combined with ALDH2 agonist improved synaptic plasticity in striatum of PD rats and delayed neurodegeneration.
