Objective: Intestinal tract is the longest digestive organ in the body.It can decompose and absorb nutrients and maintain the normal development and growth of the body.Current studies have found that small and medium intensity exercise training can be beneficial to the normal physiological function of the body's intestines and stomach.Long-term intense exercise or high-intensity exercise stress often results in impaired intestinal barrier function and disturbance of intestinal microecosystem.The gastrointestinal hormones Ghrelin,PYY,MTL and GAS secreted by the gastrointestinal tract play an important physiological role in the regulation of the gastrointestinal tract.In addition,probiotics are beneficial microorganisms for human and animal hosts,which can enhance the anti-inflammatory and anti-bacterial effects of intestinal tract,antioxidant and enhance the anti-fatigue ability of muscle.Therefore,in order to explore the changes of gastrointestinal function of athletes after probiotics intervention,we intend to establish exhaustive exercise rat model to elaborate the changes of gastrointestinal hormones in athletes.Methods: Forty SD male rats,aged 10 weeks,weighed?250 + 10?G.According to the exercise status and the supplementary dosage of rats,rats were randomly divided into quiet control group?NC?,exercise control group?ME?,quiet administration group?NCY?and exercise administration group?MEY?after one week of adaptive feeding.MEY and MEY groups were trained for 6 weeks with a speed of 19.3 m/min and a slope of 5 o.They were trained once a day for 6 days/week.In NCY and MEY groups,10.0 mL/?kg.d-1?of probiotic solution was intragastrically administered daily with a concentration of 2 g/ml.After exhaustive exercise for 6 weeks,rats were anaesthetized by intraperitoneal injection of urethane solution?20%?for 24 hours.The stomach and intestine were dissected quickly.The stomach was cut into two parts along the gastric curve.The colon was taken from the intestinal tissue.The expression of Ghrelin,PYY,MTL and GAS in the gastric and colon tissues was detected by RT-PCR and Western Blot.Results:?1?The expression of Ghrelin in gastric and intestinal tissues of rats after exhaustive exercise was higher than that of control group?P < 0.01?,but the expression of Ghrelin in gastric and intestinal tissues of rats in MEY group was higher than that in NC group?P < 0.05 or P < 0.01?,but lower than that in ME group?P < 0.05?.?2?The expression of PYY in gastric and intestinal tissues of rats after exhaustive exercise was higher than that of NC group?P < 0.01?,but the expression of PYY in gastric and intestinal tissues of NCY and MEY rats was higher than that of NC group?P 0.05?but lower than that of ME group?P < 0.05 or P < 0.01?.?3?After exhaustive exercise,the expression of MTL in gastric and intestinal tissues of rats in MEY group was lower than that in NC group?P < 0.01?,but the expression of MTL in gastric and intestinal tissues of rats in MEY group was lower than that in NC group?P 0.05?,but higher than that in ME group?P < 0.05 or P < 0.01?.The expression of MTL protein in gastric tissue of rats in MEY group was higher than that in ME group?P < 0.01?.However,the expression of MTL in intestinal tissue was lower than that in ME group?P0.05?.?4?After exhaustive exercise,the expression of GAS in gastric and intestinal tissues of rats in MEY group was lower than that in NC group?P < 0.05 or P < 0.01?,and the expression of GAS in gastric and intestinal tissues of rats in MEY group was higher than that in ME group?P < 0.05 or P < 0.01?.Conclusion:?1?The changes of Ghrelin,PYY,MTL and GAS expression in gastrointestinal tissues of rats caused by long-term exhaustive exercise are related to oxygen deficiency in rats,imbalance of energy metabolism caused by the depletion of glycogen or energy substances,and accelerated gastrointestinal emptying or gastrointestinal disorders.?2?The changes of Ghrelin,PYY,MTL and GAS expression in gastric and intestinal tissues of rats induced by long-term exhaustive exercise are closely related to the supplementation of probiotic solution.?3?Supplementation of probiotics can protect gastrointestinal mucosal cells from damage,thus maintaining the relative homeostasis of the gastrointestinal tract and preventing gastrointestinal dysfunction.
