Objective: To study the skeletal muscle metabolomics mechanism of improving depression symptoms in CUMS rats by different exercise methods based on ~1H-NMR metabolomics technology,and to provide theoretical and experimental references for the study of the “muscle-brain Crosstalk”mechanism of depression therapy.Methods: SD rats,8 weeks of age,were divided into blank control group(C),CUMS group(group D),Pre-exercise group(group P),Aerobic exercise group(group A),Resistance exercise group(group R),Aerobic+resistance Sports group(group AR).Chronic unpredictable mild stress(CUMS)was used for modeling,and the blank group was reared normally.After 4 weeks,the success of the model was verified through behavioral indicators(weight,sugar preference,and mine experiments).The model group continued to build the model to maintain depression.The rats in the exercise group were given corresponding exercise interventions while the model was created.The P and A groups used the runner treadmill for running.The resistance exercise was performed using animal weight ladders.The AR group used the above.The two exercise modes were alternated in sequence,and each group lasted 4weeks.The gastrocnemius and soleus muscles of the rats were collected the next day after the experiment,and the data were analyzed by ~1H-NMR metabolomics combined with multivariate statistics.Results:(1)Behavioral results at the end of 4 weeks of modeling showed that compared with group C,the weight,sugar and water preference rate,number of uprights,and number of crossing cells in the remaining groups were significantly lower than those in group C(p <0.05,p <0.01).),Indicating that the CUMS model was successful;after 4 weeks of exercise intervention,the behavioral indicators of rats in each experimental group were still lower than those in group C(p <0.05).Except for group C,there was no significant difference in body weight between the groups.The sugar and water preference rate and the number of uprights in group A weresignificantly higher than those in group D,and the number of crossing grids in groups A and R were significantly higher than those in group D;(2)Metabolomics results showed that compared with group C,propylene glycol in the gastrocnemius muscle of group D 4 metabolites including lactic acid,lactic acid and alanine were significantly increased(p <0.05,p <0.01);6differential metabolites including lysine,citric acid,foie gras peptide,and so on were significantly reduced(p <0.05,p <0.01).The main metabolic pathways involved are: 8 pathways including taurine and sulfite metabolism,pyruvate metabolism,and TCA cycle;group P mainly reverts 6 differential metabolites such as lactic acid and citric acid through pyruvate metabolism and TCA cycle;Group A mainly recalled 5 differential metabolites such as lactic acid and ?-glucose through pyruvate metabolism and glycolysis or gluconeogenesis;group R mainly recalled lysine and citric acid through TCA cycle and lysine biosynthesis.3 differential metabolites;AR group mainly reverted 4 differential metabolites such as alanine and citric acid through TCA cycle and alanine,aspartic acid and glutamic acid metabolism;(3)compared with group C,Four different metabolites,such as alanine,creatinine,and glycine,were significantly increased in soleus muscle of group D rats(p <0.05,p <0.01);nine different metabolites such as lactic acid,goosin,and choline were significantly reduced(p <0.05,p <0.01).The main metabolic pathways involved are: purine metabolism,glycerophospholipid metabolism and alanine,aspartic acid and glutamic acid metabolism.Group P mainly reverted 8 different metabolites such as choline and choline phosphate through glycine,serine and threonine metabolism and glycerol phospholipid metabolism;group A mainly through taurine,sulfinate metabolism and alanine,asparagus Metabolism of glutamic acid and glutamic acid significantly reverted to 8 different band metabolites such as alanine and choline;group R mainly retreated significantly through the metabolism of alanine,aspartic acid and glutamic acid,and pyruvate metabolism.5 different metabolites such as creatinine;AR group significantly reverted 5 different metabolisms such as creatinine and glutamine through D-glutamine andD-glutamic acid metabolism and alanine,aspartic acid and glutamic acid metabolism Thing.Conclusion: Different exercise methods can alleviate depressive symptoms in rats,aerobic exercise can more widely regulate the dominant metabolic pathways in different muscle fibers in skeletal muscle to show the best antidepressant effect;lactic acid metabolism may be a kind of "muscle-brain Crosstalk" Derived mechanism;metabolomics technology can provide new schemes for the mechanism research of depression therapy.
