Wheat yellow Mosaic virus(WYMV)is a member of Bymovirus,and is spreaded by Polymyxa graminis.The nuclear inclusion b(NIb)of WYMV is a protein containing a conserved Gly-AspAsp(GDD)motif,and is considered to be Rd Rp(RNA-dependent RNA polymerase)of the virus.In this study,we used WYMV NIb as a bait to capture host proteins which can interact with NIb.Subsequently,a series of molecular experiments were conducted to explore the role of NIb in the WYMV infection.The main results are as follows:At first,we obtained 12 host factors may interact with NIb through the yeast system.Consider the number of repeat selections,the Triticum aestivum light-induced protein was selected for the further investigation.Subsequently,the YTH,Bi FC and CO-IP technology are verified the vitro and vivo interaction between NIb and Ta LIP respectively.Results showed that NIb interacted with Ta LIP in vivo/vitro and this interaction affected their sub-cellular distribution.Then we took a transient silencing analysis of Ta LIP through the BSMV-induced gene silencing technology in wheat and selected the Ta LIP-silenced wheat by Fluorescence quantitative real-time PCR(q RT-PCR).Results showed that the Ta LIP-silenced wheat(BSMV: Ta LIP)showed more sensitive to WYMV.At last,we make a comparative analysis of plant hormone by LC-MS and the results showed that the ABA content of BSMV: Ta LIP was significantly reduced.In addition,we also found that external ABA Stimuli could induce the expression of Ta LIP m RNA.Finally,q RT-PCR results showed that the m RNA levels of ABA signaling pathway genes wheat abscisic acid insensitive 5(Ta ABI5),abscisic acid insensitive 8(Ta ABI8),pyrabatin resistance 1-Llike(Ta PYL1),and pyrabatin resistance3-Llike(Ta PYL3)were also suppressed in the Ta LIP-silenced wheat.In conclusion,the interaction between WYMV NIb and host Ta LIP changed their sub-cellular distribution,perturbed the ABA signaling pathway then promote the WYMV infection.
